Objective: To establish TLC scanning method for the determination of camptothecin in Camptotheca acuminata fruit, and analysis the dynamic accumulation of camptothecin in Camptotheca acuminata fruit to find out the best collection period.

Methods: Silica gel H-CMC-Na thin layer plate was adopted in the determination with chloroform-acetone (7 : 3) used as deeloper, Single-wavelength and linear scanning of TLC was used, and the detection wavelength was 360 nm.

Results: There was a good linear relationship for Comptothecin within the range of 0.0542 - 0.3252 microg, the average recovery was 97.13%, RSD was 1.76%.

Conclusion: The method is accurate, simple and reliable, and can be used for the determination of camptothecin in Camptotheca acuminata fruit and dynamic accumulation research.

Photoinduced electron transfer enables cytochrome P450 enzyme-catalyzed reaction cycling.

Cytochrome P450 enzymes (CYPs), the members of the largest superfamily of enzymes in plant kingdom, catalyze a variety of functional group transformations involved in metabolite biosynthesis, end-product derivatization, and exogeneous molecule detoxification. Nevertheless, CYPs' functional characterization and practically industrial application have been largely encumbered by their critical dependency on the reducing equivalent for the catalytic cycling, driven by the tedious electron relay mediated by CYP reductase (CPR). Here, we report a photoinduced electron transfer system that initiates and sustains the CYP-catalyzed reaction cycling.

Article Synopsis
  • The hydrolysis step of 2,7-epoxy is crucial for the production of camptothecin (CPT), a compound with potent cancer-fighting properties, yet the specific genes involved have not been previously identified.
  • This study identifies three genes (CaEH1-CaEH3) responsible for the epoxide hydrolase activity in Camptotheca acuminata and highlights their ability to catalyze the opening of various oxirane rings.
  • Functional tests show that these genes are actively involved in CPT biosynthesis, are expressed in all plant tissues with a concentration in leaves, and suggest a convergence in their evolutionary development from different ancestral genes.

Article Synopsis
  • Camptothecin (CAM) is a complex plant-derived compound known for its antitumor properties and is part of a family of monoterpene indole alkaloids (MIAs) that includes other anticancer drugs like vinblastine and vincristine.
  • Researchers have established a new in vivo enzyme assay to characterize two key enzymes involved in the biosynthesis of MIAs, focusing on the transformation of nepetalactol to 7-deoxyloganic acid, which is difficult to detect due to the instability of intermediates.
  • The study identified and characterized two cytochrome P450 enzymes and two glycosyltransferases from Camptotheca acuminata, which are responsible for key enzym

Camptothecin (CPT), an alkaloid isolated from the Camptotheca tree, has demonstrated significant anticancer properties in a range of malignancies. However, its therapeutic efficacy is limited by its hydrophobicity, poor bioavailability, and systemic toxicity. Derivatives, analogues, and nanoformulations of CPT have been synthesized to overcome these limitations.

Department of Bioengineering, School of Environmental & Biological Engineering, Nanjing University of Science & Technology, Nanjing, China.

Article Synopsis
  • A new extraction method called NADES-UAE (natural deep eutectic solvent-based ultrasound-assisted extraction) was developed to extract camptothecin (CPT) and 10-hydroxycamptothecin (10-HCPT) from fruit powder.
  • This method achieved high yields of 1.31 mg of CPT and 1.66 mg of 10-HCPT per gram of fruit, using optimal conditions such as 20% water content and a 20-minute ultrasound time.
  • The research showed that the NADES3 solvent not only improved extraction efficiency compared to traditional solvents but could also be reused multiple times, making it a sustainable and eco-friendly option for extraction processes.
