We show that for the system of SH-containing compounds of the organism (cysteine, in particular) there are conditions leading to immunosuppression, which occurs when the level of amino acid cysteine in blood serum increases. The arising "overload" of free sulfhydryl groups inactivates antibodies of the IgM class of any specificity restoring disulphide intramolecular bonds and then the number of immune antibody-producing cells, mitotic activity of T- and B-lymphocytes and interleukin-2 synthesis are lowered and the mobility of surface immunoglobulin B lymphocytes receptors is damaged. Determination of total blood sulfhydryl groups for diagnostics of developing immunodeficiency disorder is not informative because of the high individual fluctuation of blood sulfhydryl groups number. It is the determination of inhibitory properties of blood serum regarding to IgM antibodies of any class and specificity in test system in vitro that is informative. It is proved clinically and by experiments that in the course of infectious inflammatory process the long-lasting inhibitory properties of blood serum are unfavorable and lead to the lingering course of disease. On the contrary, in case of autoimmune pathology the appearance of inhibitory properties of blood serum is a favorable prognostic factor.

A π-Extended AIE Platform for Pattern Recognition of Glycosaminoglycans.

Anal Chem

Department of Chemistry, College of Sciences, Nanjing Agricultural University, Nanjing, Jiangsu 210095, People's Republic of China.

Accurate discrimination of complicated glycosaminoglycans is a challenging but meaningful task for ensuring their safe use in clinics. With the purpose of reducing the production cost of sensor arrays for glycosaminoglycans, three fluorescence turn-on sensors named , , and were readily synthesized by simple alkylation of the pyridyl units of the π-extended AIEgen, namely, tetra-(4-pyridylphenyl) ethylene. The designed sensors are cross-reactive toward tested glycosaminoglycans including heparin, chondroitin sulfate, hyaluronic acid, and dextran sulfate, whose mechanism could be ascribed to the multivalent electrostatic, CH···π, and hydrophobic interactions between the sensors and different glycosaminoglycans to form corresponding fluorescent aggregates.

Serial Total Bile Acid Measurements in Intrahepatic Cholestasis of Pregnancy.

Although peak serum total bile acid (TBA) levels guide management of intrahepatic cholestasis of pregnancy (ICP), whether ICP progresses in severity and when or how to assess bile acid levels serially remains unclear. We conducted a secondary analysis of a single-institution retrospective cohort study to assess bile acid trends across pregnancy among individuals diagnosed with ICP and to evaluate whether there was progression to higher ICP severity. We defined ICP severity as mild (peak TBA less than 40 micromol/L), moderate (peak TBA between 40 and 100 micromol/L), or severe (peak TBA 100 micromol/L or greater).

Effects of Chemical Pretreatments of Wood Cellulose Nanofibrils on Protein Adsorption and Biological Outcomes.

ACS Appl Mater Interfaces

January 2025

Center of Translational Oral Research (TOR), Department of Clinical Dentistry, University of Bergen, Bergen 5009, Norway.

Wood-based nanocellulose is emerging as a promising nanomaterial in the field of tissue engineering due to its unique properties and versatile applications. Previously, we used TEMPO-mediated oxidation (TO) and carboxymethylation (CM) as chemical pretreatments prior to mechanical fibrillation of wood-based cellulose nanofibrils (CNFs) to produce scaffolds with different surface chemistries. The aim of the current study was to evaluate the effects of these chemical pretreatments on serum protein adsorption on 2D and 3D configurations of TO-CNF and CM-CNF and then to investigate their effects on cell adhesion, spreading, inflammatory mediator production , and the development of foreign body reaction (FBR) .

A novel ionic liquid MALDI matrix, 3-aminoquinoline/2',4',6'-trihydroxyacetophenone monohydrate (3-AQ/THAP), was developed for the rapid qualitative and quantitative detection of miRNA from biological samples. Compared to the traditional matrix 2,5-dihydroxybenzoic acid (DHB) and previously reported oligonucleotide-specific matrices, such as 3-aminopicolinic acid (3-APA), 3-hydroxypicolinic acid (3-HPA), and 6-aza-2-thiothymine (ATT), the 3-AQ/THAP matrix offers several advantages. It produces fewer alkali metal adduct peaks, exhibits higher sensitivity, and ensures better spot-to-spot repeatability.

Homeobox protein MSX-1 restricts hepatitis B virus by promoting ubiquitin-independent proteasomal degradation of HBx protein.

Hepatitis B virus (HBV) X protein (HBx) is a key factor for regulating viral transcription and replication. We recently characterized homeobox protein MSX-1 (MSX1) as a host restriction factor that inhibits HBV gene expression and genome replication by directly binding to HBV enhancer II/core promoter (EnII/Cp) and suppressing its promoter and enhancer activities. Notably, HBx expression was observed to be repressed more drastically by MSX1 compared to other viral antigens.
