The aqueous self-assembly of oligopeptide-flanked pi-conjugated molecules into discrete one-dimensional nanostructures is described. Unique to these molecules is the fact that the pi-conjugated unit has been directly embedded within the peptide backbone by way of a synthetic amino acid with pi-functionality that is compatible with standard Fmoc-based peptide synthesis. The peptide-based molecular design enforces intimate pi-pi communication within the aggregate after charge-screening and self-assembly, making these nanostructures attractive for optical or electronic applications in biological environments. The synthesis and assembly are reported along with spectroscopic and morphological characterization of the new nanomaterials.

Combining polysaccharides with polypeptides enables growth of diverse nanostructures with minimal toxicity, low immune response, and potential biodegradability. However, examples of nanostructures combining polysaccharides with polypeptides are limited due to synthetic difficulties and related issues of solubility, purification, and characterization, with previous reports of polysaccharide-block-polypeptide block copolymers requiring methods such as polymer-polymer coupling and post-polymerization modifications paired with difficult purification steps. Here, we synthesized dextran-block-poly(benzyl glutamate) block copolymers in water via polymerization-induced self-assembly (PISA) to form nanostructures in situ, studying their morphologies using experimental methods and molecular modeling.

This study aims to explore the development of natural bio-based amphiphilic block copolymers for drug delivery applications. We investigated block copolymers derived from tamarind seed xyloglucan and solanesol, focusing on their synthesis, structural analysis, aqueous self-assembly, and drug encapsulation. Specifically, xyloglucan hydrolysate segments with number-average degrees of polymerization (DPs) of between 8 and 44 (XOS, XMS, XMS, XMS, and XMS) were used as the hydrophilic blocks, whereas plant-sourced solanesol was selected as the hydrophobic segment.
