A method to obtain (Z)-beta,gamma-unsaturated nitriles in high or good yields and with moderate or high stereoselectivity is described. This report explores the potential of novel 6-aryloxy-2-aminopyrimidine-benzonitrile scaffolds as promising anti-infective agents in the face of the increasing threat of infectious diseases. Starting from 2-amino-4,6-dichloropyrimidine, a series of 24 compounds inspired from the antiviral drugs dapivirine, etravirine, and rilpivirine were designed and synthesized via a two-step reaction sequence in good yields. Two new quinazolinone nitriles (1 and 2) and one new indole alkaloid (3), together with 13 known compounds, were isolated from the deep-sea-derived MCCC 3A00265. Their structures were determined by extensive spectroscopic analysis, with the absolute configurations established by comparing experimental and calculated electronic circular dichroism (ECD) and optical rotation (OR) data as well as biogenetic considerations. Viricyanoamides A and B (1 and 2) are the sole representatives of quinazolinones featuring a nitrile group, while solitumidine F (3) incorporates a rare pyrrolidinedione unit as an indole terpenoid.

This study investigates the mechanism of prototropic tautomerization in metal-bound asymmetric pyrazole (R-PzH) ligands during Cu(II)-mediated PzH-MeCN coupling reactions. Intrinsic prototropic tautomerization of metal-bound ligands has not been previously documented. Various new bis-pyrazolylamidino Cu(II) complexes, [Cu(R-Pz(HNC(Me)))(ClO)], from the coupling reaction, and tetrakis pyrazole Cu(II) complexes, [Cu(R-PzH)(ClO)], with symmetric and asymmetric -monosubstituted R-PzH ligands were synthesized and characterized.
