M1 aminopeptidases comprise a large family of biologically important zinc enzymes. We show that peptide turnover by the M1 prototype, leukotriene A4 hydrolase/aminopeptidase, involves a shift in substrate position associated with exchange of zinc coordinating groups, while maintaining the overall coordination geometry. The transition state is stabilized by residues conserved among M1 members and in the final reaction step, Glu-296 of the canonical zinc binding HEXXH motif shuffles a proton from the hydrolytic water to the leaving group. Tripeptide substrates bind along the conserved GXMEN motif, precisely occupying the distance between Glu-271 and Arg-563, whereas the Arg specificity is governed by a narrow S1 pocket capped with Asp-375. Our data provide detailed insights to the active site chemistry of M1 aminopeptidases and will aid in the development of novel enzyme inhibitors.

Background: Cardiovascular disease (CVD) remains the leading cause of death in pregnant and peripartal women in western countries. Physiological changes during pregnancy can lead to cardiovascular complications in the mother; women with pre-existing heart disease may not tolerate these changes well, increasing their susceptibility to adverse cardiovascular outcomes during pregnancy. CO hydrogenation to methanol using green hydrogen derived from renewable resources provides a promising method for sustainable carbon cycle but suffers from high selectivity towards byproduct CO. Here, we develop an efficient PdZn-ZnO/TiO catalyst by engineering lattice dislocation structures of TiO support. We discover that this modification orders irregularly arranged atoms in TiO to stabilize crystal lattice, and consequently weakens electronic interactions with supported active phases.

Cycloparaphenylenes (CPPs) represent a significant challenge for the synthesis of mechanically interlocked architectures, because they lack heteroatoms, which precludes traditional active and passive template methods. To circumvent this problem and explore the fundamental and functional properties of CPP rotaxanes and catenanes, researches have resorted to unusual non-covalent and even to labor-intensive covalent template approaches. Herein, we report a ring-in-ring non-covalent template strategy that makes use of the surprisingly strong non-covalent inclusion of crown ethers into suitably sized CPPs.

The stable operation of high-capacity lithium-sulfur batteries (LSBs) has been hampered by slow conversion kinetics of lithium polysulfides (LiPSs) and instability of the lithium metal anodes. Herein, 6-(dibutylamino)-1,3,5-triazine-2,4-thiol (DTD) is introduced as a functional additive for accelerating the kinetics of cathodic conversion and modulating the anode interface. We proposed that a coordination interaction mechanism drives the polysulfide conversion and modulates the Li solvated structure during the binding of the N-active site of DTD to LiPSs and lithium salts.
