Data mining by pairwise comparison of over 150,000 human liver microsome (HLM) intrinsic clearance values stored within the internal Pfizer database has been performed by an automated tool. Systematic probability tables of specific structural changes on the intrinsic clearance of phenyl derivatives have been generated. From these data two new parameters, the Pfizer Metabolism Index (PMI) and Metabolism-Lipophilicity Efficiency (MLE) are introduced for each fragment. The findings are applied to a Topliss style analysis that focuses on metabolic stability.

Unsymmetrical bis-cyclometalated dicarboxylato complexes (-6-32)-[Pt(tpy)(OCR)] [tpy = cyclometalated 2-(-tolyl)pyridine, R = -Bu (), Me (), Ph (), CF ()], are obtained from the reaction of -[Pt(tpy)] with the appropriate PhI(OCR) reagent. Treatment of complexes of this type with different carboxylates (R'CO) results in the formation of mixed-carboxylato derivatives, namely (-6-43)-[Pt(tpy)(OCMe)(OCR')] [R' = -Bu (), CF (), Ph ()], (-6-34)-[Pt(tpy)(OCCF)(OCR')] [R' = -Bu (), Me (), Ph ()], and (-6-34)-[Pt(tpy)(OC--Bu)(OCMe)] (). Irradiation of - and - with UV light (365 nm) in MeCN gives 5-methyl-2-(2-pyridyl)phenyl pivalate (), 5-methyl-2-(2-pyridyl)phenyl acetate () or 5-methyl-2-(2-pyridyl)phenyl benzoate () as the major photoproduct from most complexes, resulting from a reductive C-O coupling between a tpy ligand and a carboxylato ligand.

In this work, inspired by a facile strategy in point-of-care detection, two devices were fabricated to detect small molecules via photocurrent measurement. A linear response of the photocurrent against the concentration of the small molecules was found.

Synthesis of Atropisomeric Quinazolin-4-one Derivatives Based on Remote H/D and C/C Discrimination.

Both enantiomers of 2-ethylquinazolin-4-ones bearing -CHO/CDO and CHO/CHO phenyl groups at the N3 position were prepared. These are isotopic atropisomeric compounds based on a remote and conformationally flexible H/D and C/C discrimination, and it was found that a CHCl solution of -CHO/CDO derivative shows a slight specific optical rotation. Furthermore, diastereomeric quinazolinone derivatives bearing a chiral carbon were prepared, and their stereochemical purities and rotational stability as well as the isotopic atropisomerism were verified by H NMR and chiral high-performance liquid chromatography (HPLC) analyses.

Suitable structural modifications of the functional groups at N-substituent of (-)-cis-N-normetazocine nucleus modulate the affinity and activity profile of related ligands toward opioid receptors. Our research group has developed several compounds and the most interesting ligands, LP1 and LP2, exhibited a dual-target profile for mu-opioid receptor (MOR) and delta-opioid receptor (DOR). Recent structure-affinity relationship studies led to the discovery of novel LP2 analogs (compounds 1 and 2), which demonstrated high MOR affinity in the nanomolar range.

This paper presents a copper(I)-catalyzed intramolecular tandem acylation/-arylation of methyl 2-[2-(2-bromophenyl)acetamido]benzoates for the synthesis of benzofuro[3,2-]quinolin-6(5)-ones under mild conditions. The combination of CuI, 1,10-phenanthroline, and KCO in DMSO was found to be the optimal reaction condition, producing the target products in high yields (84-99%) at 70 °C for 16 h. The tandem reaction was applicable to substrates bearing halo, electron-withdrawing, and electron-donating groups at their phenyl moieties with a broad substrate scope.
