A guide to analysing Universal Eating Monitor data: assessing the impact of different analysis techniques.

Cumulative intake curves and within-meal changes in subjective feelings of fullness can provide researchers with detailed data on the effects of psychological, nutritional or pharmacological manipulations on the expression of human appetite. However, a number of different approaches to the collection and analysis of within-meal data exist resulting in potential to produce contrasting findings. The current study measured cumulative intake and change in appetite using a Universal Eating Monitor (UEM). Three different techniques (area under the curve, visual ascription, and a coefficient approach) were used to analyse the same cumulative intake curves produced in a study of stress on food intake. Twenty-three adult participants (mean age 21 years) consumed a meal comprised of pasta and marinara sauce and, with the aid of the Sussex Meal Pattern Monitor (SMPM), were periodically interrupted to measure subjective feelings of fullness. As hypothesised, analysing cumulative intake curves with differing techniques affected the overall study findings. No significant between-condition differences in the cumulative intake or fullness curves were found using either the visual ascription or the area under the curve approaches. In contrast, the coefficient approach found a significant difference in the fullness curves between relaxation and cold pressor conditions (p=0.012). This discrepancy in findings was due to the presence of a quadratic component in the cumulative intake curve in the stress condition which was not present in control (p=0.017). Whilst the relative merits of various approaches to microstructural analysis of eating behaviour remain to be fully evaluated, the case for some form of standardised analytic approach may need to be addressed.

Introduction: Fluid overload (FO), a state of pathologic positive cumulative fluid balance (CFB), is common in Pediatric Intensive Care Units (PICU) and associated with morbidity and mortality. Because different PICUs may have unique needs, barriers, and limitations to accurately report fluid balance (FB) and reduce FO, understanding the drivers of positive FB is needed. We hypothesize CFB >5% and >10% is common within initial days of PICU admission, but that reasons for high %CFB will vary across sites, as will barriers to accurate FB recording and opportunities to improve FB recording and management.

The toxicokinetics of nitrosamines remain a mystery to this day, though it appears that the role of nitrosamines in potentiating the generation of mutations required for the onset of skin cancer continues to be a significant concern. Nitrosamines are mutagens, genotoxic substances, and mediators of phototoxicity/carcinogenicity, whose long-term daily usage, in the context of polypharmacy, can result in the parallel appearance of heterogeneous forms of skin cancer: keratinocytic and melanocytic. But a number of clinical observations suggest that it is the nitrosamines that potentiate the multiple occurrences of skin cancer over the years, or recurrences of skin cancer localized in areas exposed to solar radiation.

We recently reported that a chimeric peptide (GEP44) targeting the glucagon-like peptide-1 receptor (GLP-1R) and neuropeptide Y1- and Y2-receptors decreased body weight (BW), energy intake and core temperature in diet-induced obese (DIO) male and female mice. Given that GEP44 was found to reduce core temperature (surrogate measure of energy expenditure (EE)) in DIO mice, we hypothesized that GEP44 would reduce EE in male and female high fat diet (HFD)-fed rats. To test this, rats were maintained on a HFD for at least 4 months to elicit DIO prior to undergoing a sequential 2-day vehicle period, 2-day GEP44 (50 nmol/kg) period and a minimum 2-day washout period and detailed measures of energy homeostasis.
