Reactions of substituted acetone derivatives with acrylic acid esters (>200 mol %) in the presence of t-BuOK (200 mol %) in t-BuOH-THF (1:1 by volume) turned out to proceed as a cascade process consisting of the first Michael addition, the second Michael addition, and the last Claisen reaction to afford 4,4-disubstituted cyclohexane-1,3-diones. Only more substituted enolates play the role of a Michael donor in this cascade process, and therefore the ketone took up two alkoxycarbonylethyl groups on the same carbon bearing more substituents. Such intermediates were followed by intramolecular Claisen reactions leading to cyclohexane-1,3-diones bearing quaternary stereogenic centers at C(4), which bears an alkoxycarbonylethyl group and the substituent of the starting acetone derivatives. Thus-obtained 4,4-disubstituted cyclohexane-1,3-diones were successfully employed for total syntheses of intricate alkaloids of biological interest such as (+)-aspidospermidine, (+/-)-galanthamine, (+/-)-lycoramine, and (+/-)-mesembrine, all featuring quaternary stereogenic centers. DFT calculations provided us with clear-cut explanations for the observed chemoselectivity of the cascade process involving ketone-based enolates under thermodynamically controlled conditions.

Nanozymes have shown significant potential in cancer catalytic therapy by strategically catalyzing tumor-associated substances and metabolites into toxic reactive oxygen species (ROS) , thereby inducing oxidative stress and promoting cancer cell death. However, within the complex tumor microenvironment (TME), the rational design of nanozymes and factors like activity, reaction substrates, and the TME itself significantly influence the efficiency of ROS generation. To address these limitations, recent research has focused on exploring the factors that affect activity and developing nanozyme-based cascade catalytic systems, which can trigger two or more cascade catalytic processes within tumors, thereby producing more therapeutic substances and achieving efficient and stable cancer therapy with minimal side effects.

Introduction: Current models of Alzheimer's disease (AD) progression assume a common pattern and pathology, oversimplifying the heterogeneity of clinical AD.

Methods: We define a syndrome as a unique biomarker progression pattern and develop a lag measure to cluster pre-dementia individuals, reflecting their pathology's multi-dimensionality. A novel HSO-catalyzed ANRORC-type rearrangement of pyrazinones to imidazoles proceeding through pyridazino[]annulation with simultaneous introduction of a pyrazole ring at position 2 of the imidazole system has been developed, which offers efficient and expedited access to new biheterocyclic systems - 2-(pyrazol-3-ul)imidazoles and 2-(pyrazol-3-yl)imidazo[4,5-]pyridazines. Diverse bi--heterocyclic systems with the imidazo[4,5-]pyridazine-4,7-diamine moiety could be obtained in excellent yield when 5,6-dicyano-3-(2-oxo-2-ethyl)pyrazin-2(1)-ones interact with hydrazines the selective spiro-formation in a tandem ring-opening/ring-closing process, which allowed the simultaneous construction of five new C-N bonds. This new method is compatible with an array of functional groups, proceeds under mild reaction conditions with the involvement of commercially available reagents.
