Cross-regulatory mechanisms in hormone signaling.

Published: March 2009

Recent studies suggest that hormones act through a web of interacting responses rather than through isolated linear pathways. This signal integration architecture may be one mechanism for increasing the specificity of outcomes in different cellular contexts. Several common themes for cross-regulation between pathways can be observed. Here, we propose a classification scheme for different levels of signaling pathway cross-regulation. This scheme is based on which parts of the individual pathways are acting as information conduits between pathways. Examples from the recent plant hormone biology literature are used to illustrate the different modes of interaction.

N6 adenosine and C5 cytosine modification of mRNAs, tRNAs and rRNAs are regulated by the behaviour of distinct sets of writer, reader and eraser effector proteins which are conventionally considered to function independently. Here, we provide evidence of global cross-regulatory and functional interaction between the mA and mC RNA methylation systems. We first show that mA and mC effector protein transcripts are subject to reciprocal base modification supporting the existence of co-regulatory post-transcriptional feedback loops.

Ischemic stroke, as one of the most severe and prevalent neurological disorders, poses a significant threat to the health and quality of life of affected individuals. Stemming from the obstruction of blood flow, ischemic stroke, leads to cerebral tissue hypoxia and ischemia, instigating a cascade of pathophysiological changes that markedly exacerbate neuronal damage and may even culminate in cell death. In recent years, emerging research has increasingly focused on novel cell death mechanisms such as ferroptosis and cuproptosis.

Article Synopsis
  • Gouty arthritis (GA) is mainly a joint disease linked to metabolic and immune dysfunctions, with dysbiosis (imbalance) of gut bacteria recognized as a key contributing factor.
  • Changes in gut flora may trigger metabolic disorders that lead to GA, particularly in individuals with high uric acid levels, but the specific mechanisms remain unclear.
  • Therapies for GA can influence gut bacteria, indicating that managing gut flora could provide new ways to prevent and treat GA, with potential insights from traditional Chinese medicine for lowering uric acid and alleviating symptoms.

Metabolic Responses to Redox Stress in Vascular Cells.

Antioxid Redox Signal

November 2024

Division of Cardiovascular Medicine, Department of Medicine, Brigham and Women's Hospital and Harvard Medical School, Boston, Massachusetts, USA.

Redox stress underlies numerous vascular disease mechanisms. Metabolic adaptability is essential for vascular cells to preserve energy and redox homeostasis. Single-cell technologies and multiomic studies demonstrate significant metabolic heterogeneity among vascular cells in health and disease.

Unproductive splicing is a mechanism of post-transcriptional gene expression control in which premature stop codons are inserted into protein-coding transcripts as a result of regulated alternative splicing, leading to their degradation via the nonsense-mediated decay pathway. This mechanism is especially characteristic of RNA-binding proteins, which regulate each other's expression levels and those of other genes in multiple auto- and cross-regulatory loops. Deregulation of unproductive splicing is a cause of serious human diseases, including cancers, and is increasingly being considered as a prominent therapeutic target.
