Objective: To compare the development and implantation of human embryos biopsied with two different methods for preimplantation genetic diagnosis (PGD).

Design: Technique and method.

Setting: A regional hospital IVF laboratory and private reproductive medicine clinic.

Patient(s): Women undergoing IVF and PGD.

Intervention(s): Day 3 embryos were biopsied with aspiration and displacement; the embryos were cultured to blastocyst stage and then transferred.

Main Outcome Measure(s): Blastocyst rate, pregnancy rate, and implantation rate.

Result(s): One hundred fifty-one embryos from 14 patients were biopsied with the blastomere displacement method and 51 embryos from 5 patients were biopsied with the aspiration method. Displacement used less time than aspiration; thus, the time of embryos exposed to biopsy solution was shorter when displacement was used. Blastocyst formation (55.6%-56.8%) and ongoing pregnancy rate (50%) were not different between the two biopsy methods. However, the implantation rate was significantly higher in patients with embryos biopsied using the displacement method (64.7%) than with the aspiration method (25%).

Conclusion(s): Blastomere displacement uses less time and is an easy and simple method for embryo biopsy and could be used as an alternative method for embryo biopsy. Our results indicate that the displacement method minimizes embryo damage during biopsy that was indicated by a higher implantation rate.

Modulation of placental angiogenesis by metformin in a rat model of gestational diabetes.

Histochem Cell Biol

Medical Histology and Cell Biology Department, Faculty of Medicine, Mansoura University, Mansoura, 35516, Egypt.

Gestational diabetes mellitus (GDM) significantly disrupts placental structure and function, leading to complications such as intrauterine growth restriction (IUGR) and preeclampsia. This study aimed to investigate the effects of GDM on placental histology, angiogenesis, and oxidative stress, as well as evaluate metformin's protective role in mitigating these changes. A total of 60 pregnant Sprague-Dawley rats were divided into four groups: control, metformin-treated, GDM, and GDM with metformin.

Correlation among blastocoel fluid DNA level, apoptotic genes expression and preimplantation aneuploidy.

It is believed that aneuploid embryos release cell-free DNA (cfDNA) into the blastocyst cavity during the self-correction process through the apoptotic mechanism. This study aimed to develop less invasive methods for predicting ploidy status by investigating how ploidy status affects blastocoel fluid DNA (BF-DNA) levels and apoptotic gene expression as indicators of embryo viability. Human blastocysts were classified into three groups; Survivable Embryo (SE), Fatal Single and double Aneuploidy (FSDA), and Multiple Aneuploidy (MA) using array comparative genomic hybridization (array-CGH) by trophectoderm (TE) biopsy.

Background: Embryo selection for transfer is critical in assisted reproduction. The presence of DNA in the blastocoel cavity of human blastocysts is assumed to be a consequence of common preimplantation chromosomal abnormalities.

Objective: This study examined the relationship between the amount of blastocoel fluid (BF) DNA and the band intensity of amplified BF-DNA in gel electrophoresis, considering the influence of ploidy status.

Fistulization involving both the sigmoid colon and urachus is exceedingly rare. While previous cases have often necessitated laparotomy due to the involvement of multiple organs, only one instance of successful laparoscopic surgery has been reported. Here, we present the second documented case of laparoscopic resection of a sigmoid-urachal fistula.

Background And Aims: Maternal obesity increases the risk of the paediatric form of metabolic dysfunction-associated steatotic liver disease (MASLD), affecting up to 30% of youth, but the developmental origins remain poorly understood.

Methods: Using a Japanese macaque model, we investigated the impact of maternal Western-style diet (mWSD) or chow diet followed by postweaning WSD (pwWSD) or chow diet focusing on bile acid (BA) homeostasis and hepatic fibrosis in livers from third-trimester fetuses and 3-year-old juvenile offspring.

Results: Juveniles exposed to mWSD had increased hepatic collagen I/III content and stellate cell activation in portal regions.
