Effects of chloroquine and primaquine on rat liver cytosolic N-acetyltransferase activity.

Published: July 1991

Chloroquine caused only slight reductions in NAT activity when added in vitro, and had no detectable influence when animals were pretreated with it for 4 days. This would suggest that the previously reported reduced excretion of acetylated metabolites of INH and SDD following chloroquine pretreatment is not the result of inhibition of NAT. In contrast, we found that primaquine significantly (P less than 0.05) reduced NAT activity when added in vitro, suggesting the need for further study with this agent.

Article Synopsis
  • A new species of Pleosporales fungus was isolated from driftwood and cultivated to explore potential new secondary metabolites.
  • The study identified eight new compounds called ophiobolin-type sesterterpenes, known as bipolarolides H-O, using a combination of HPLC-MS and spectroscopic methods.
  • Three of these bipolarolides demonstrated inhibitory activity against certain organisms, with MIC values indicating their potency.

From the leaves of , fourteen compounds were isolated and identified: D-mannitol (), a mixture of β-sitosterol () and stigmasterol (), α-amyrin (), betulin (), lupeol (), lupenone (), betulinic acid (), taraxerol (), 3β-(E)-coumaroyltaraxerol (), 3β-(Z)-coumaroyltaraxerol (), ursolic acid (), stigmasterol 3-O-β-D-glucoside (), and β-sitosterol 3-O-β-D-glucoside (). These compounds were analysed through NMR spectroscopy (both 1D and 2D) and by comparing them to previously published data. Compounds , , , and - have been identified from this species for the first time.

ADSL promotes autophagy and tumor growth through fumarate-mediated Beclin1 dimethylation.

Zhejiang Provincial Key Laboratory of Pancreatic Disease, The First Affiliated Hospital, Institute of Translational Medicine, Zhejiang University School of Medicine, Zhejiang University, Hangzhou, China.

As an enzyme with a critical role in de novo purine synthesis, adenylosuccinate lyase (ADSL) expression is upregulated in various malignancies. However, whether ADSL possesses noncanonical functions that contribute to cancer progression remains poorly understood. Here, we demonstrate that protein kinase R-like endoplasmic reticulum kinase (PERK) activated by lipid deprivation or ER stress phosphorylates ADSL at S140, leading to an enhanced association between ADSL and Beclin1.

Oligodendrocytes in Alzheimer's disease pathophysiology.

Nat Neurosci

January 2025

Institute of Neuronal Cell Biology, Technical University Munich, Munich, Germany.

Our understanding of Alzheimer's disease (AD) has transformed from a purely neuronal perspective to one that acknowledges the involvement of glial cells. Despite remarkable progress in unraveling the biology of microglia, astrocytes and vascular elements, the exploration of oligodendrocytes in AD is still in its early stages. Contrary to the traditional notion of oligodendrocytes as passive bystanders in AD pathology, emerging evidence indicates their active participation in and reaction to amyloid and tau pathology.
