The reaction of the phosphetane disulfide, FcP(S)S 2P(S)Fc ( 1) (Fc = (eta (5)-C 5H 5)Fe(eta (5)-C 5H 4)), the ferrocenyl analogue of the Lawesson reagent, with gold and palladium complexes leads to the unprecedented formation of phosphonodithioate ligands upon coordination to the metal centers. The reaction of 1 with gold complexes such as [AuCl(PR 3)] affords the species [Au{S 2P(OH)Fc}(PR 3)] (PR 3 = PPh 3 ( 2), PPh 2Me ( 3)), in which the phosphonodithioate ligand Fc(OH)PS 2 (-) has been formed. The same ligand is present in the compound [Au 2{S 2P(OH)Fc} 2].[N(PPh 3) 2]Cl ( 4), obtained by reaction of 1 with [N(PPh 3) 2][AuCl 2]. It crystallizes with one molecule of [N(PPh 3) 2]Cl, whereby complex 4 acts as an anion receptor and forms strong hydrogen bonds between the chloro and the hydroxyl groups. The reaction with palladium derivatives is different; two complexes, [Pd 2(S 4OP 2Fc 2) 2] ( 5) and [Pd 4Cl 4(S 4OP 2Fc 2) 2] ( 6), are obtained in molar ratio 2:1 and 1:1, respectively. In these complexes a new phosphonodithioate ligand is present and probably arises from the condensation of two molecules of Fc(OH)PS 2 (-). Complex 5 has also been characterized by X-ray methods.

Cancer immunotherapies rely on CD8 cytolytic T lymphocytes (CTLs) in recognition and eradication of tumor cells via antigens presented on major histocompatibility complex class I (MHC-I) molecules. However, we observe MHC-I deficiency in human and murine urologic tumors, posing daunting challenges for successful immunotherapy. We herein report an unprecedented nanosonosensitizer of one-dimensional bamboo-like multisegmented manganese dioxide@manganese-bismuth vanadate (BMMBV) to boost multiple branches of immune responses targeting MHC-I-deficient tumors.

Helicase is a nucleic acid motor that catalyses the unwinding of double-stranded (ds) RNA and DNA via ATP hydrolysis. Helicases can act either as a nucleic acid motor that unwinds its ds substrates or as a chaperone that alters the stability of its substrates, but the two activities have not yet been reported to act simultaneously. Here, we used single-molecule techniques to unravel the synergistic coordination of helicase and chaperone activities, and found that the severe acute respiratory syndrome coronavirus helicase (nsp13) is capable of two modes of action: (i) binding of nsp13 in tandem with the fork junction of the substrate mechanically unwinds the substrate by an ATP-driven synchronous power stroke; and (ii) free nsp13, which is not bound to the substrate but complexed with ADP in solution, destabilizes the substrate through collisions between transient binding and unbinding events with unprecedented melting capability.

Alzheimer's disease (AD) is a debilitating neurodegenerative disease that is marked by profound neurovascular dysfunction and significant cell-specific alterations in the brain vasculature. Recent advances in high throughput single-cell transcriptomics technology have enabled the study of the human brain vasculature at an unprecedented depth. Additionally, the understudied niche of cerebrovascular cells, such as endothelial and mural cells, and their subtypes have been scrutinized for understanding cellular and transcriptional heterogeneity in AD.
