Introduction: Granulysin is a cytotoxic molecule involved in cellular immune reactions.

Materials And Methods: The levels of granulysin mRNA in the peripheral blood and serum granulysin of patients with primary biliary cirrhosis (PBC) were determined by quantitative polymerase chain reaction and enzyme-linked immunosorbent assay, respectively. The expression of granulysin mRNA and serum protein in PBC was increased compared to the controls.

Results: The expression of granulysin mRNA or serum protein showed close associations, respectively, with natural killer cell population in PBC patients. Serum granulysin was down-regulated by steroid and ursodeoxycholic therapy for PBC according to the improvement of severity of PBC. In addition, the expression of serum granulysin were related to serum total bilirubin and Mayo Clinic risk score. The serum granulysin reflected the cellular immune status of patients with PBC, and the expression correlated with the severity of PBC.

Conclusion: Therefore, there is a clinical benefit to monitoring granulysin as a biomarker of the prognosis of patients with PBC.

Article Synopsis
  • Endometriosis is a chronic condition where tissue similar to the uterus grows outside it, and this study looks at immune molecules and their connection to infertility in women with endometriosis.
  • Researchers compared 87 infertile women, finding higher levels of PRF1 and MICA in those with endometriosis, especially in more severe cases.
  • The study suggests that these molecules could serve as potential biomarkers for endometriosis severity and advocates for further research on their therapeutic implications.

Article Synopsis
  • - This study investigates genetic variations in the GNLY and PTX3 genes and their link to acne in Egyptian individuals, highlighting the inflammatory nature of acne as a skin disorder.
  • - The research included 180 acne patients and 180 healthy controls, using real-time PCR to analyze specific genetic polymorphisms (SNPs) related to these genes.
  • - Results showed that the PTX3 rs2305619 variant is significantly associated with acne susceptibility, indicating that individuals with certain genotypes (AG + GG) are more likely to develop acne, while the GNLY rs7908 polymorphism does not show a similar association.

Efficacy of NKG2D CAR-T cells with IL-15/IL-15Rα signaling for treating Epstein-Barr virus-associated lymphoproliferative disorder.

Epstein-Barr virus (EBV) related post-transplant lymphoproliferative disorder (EBV-PTLD) is a life-threatening complication after hematopoietic stem cell transplantation (HSCT) or solid organ transplantation (SOT), for which no standard therapeutic means have been developed. Significant increase expression of natural killer group 2 member D ligands (NKG2DLs) was observed on B-lymphoblastoid cells of EBV-PTLD, indicating NKG2DLs as potential therapeutic targets for treatment of EBV-PTLD. In this study, the recombinant constructs of NKG2D CAR and IL-15/IL-15Rα-NKG2D CAR were generated with a retroviral vector and then transduced to human T cells to produce NKG2D CAR-T and IL-15/IL-15Rα-NKG2D CAR-T cells, respectively.

Serum Granulysin as a Possible Key Marker of Vitiligo Activity and Severity.

Indian Dermatol Online J

April 2024

Department of Dermatology, Venereology, and Andrology, Faculty of Medicine, Benha University, Benha, Egypt.

Background: Vitiligo is an immune-mediated, chronic skin condition that affects both the innate and adaptive immune systems. Antimicrobial peptide overexpression is one of its defining characteristics. Granulysin (GNLY), an antimicrobial peptide, may play a role in the pathogenesis of various autoimmune diseases.

Assessment of serum granulysin and cathepsin-L levels in vitiligo patients.

Rev Assoc Med Bras (1992)

May 2024

Gaziosmanpasa University, School of Medicine, Department of Dermatology and Venereology - Tokat, Turkey.

Objective: Cellular and humoral immunity plays a role in the pathogenesis of vitiligo. T lymphocytes and natural killer cells involved in cellular immunity carry out their cytotoxic activities through perforin/granzyme-dependent granule exocytosis, in which granulysin and cathepsin-L are also involved. The aim of this study was to investigate the possible role of serum granulysin and cathepsin-L in the etiopathogenesis of vitiligo and their association with disease activity and severity.
