Prussian Blue Dispersed Sphere Catalytic Labels for Amplified Electronic Detection of DNA.

A highly sensitive electrochemical DNA hybridization assay using Prussian blue (PB)-modified polymeric spheres as the oligonculeotide labeling tag is described. The sandwich assay relies on a secondary nucleic-acid probe functionalized with polystyrene beads loaded with numerous Prussian blue nanoparticles. The very strong catalytic activity of the captured PB 'artificial peroxidase' tag towards the reduction of hydrogen peroxide, along with the encapsulation of numerous catalytic particles onto polymeric beads, allows amperometric detection of the DNA target down to the 50 fM level (2.5 amol). Imaging and spectroscopic measurements are used to characterize the PB-tagged polystyrene beads. Such coupling of PB catalytic labels with polymeric carrier beads offers great promise for amplified transduction of different biomolecular interactions.

Prussian blue nanoparticles (PBNPs) have been identified as a promising candidate for biomimetic peroxidase (POD)-like activity, specifically due to the metal centres (Fe/Fe) of Prussian blue (PB), which have the potential to function as catalytically active centres. The decoration of PBNPs with desired functional polymers (such as amino- or carboxylate-based) primarily facilitates the subsequent linkage of biomolecules to the nanoparticles for their use in biosensor applications. The growing market for sodium-ion batteries has stimulated interest in research on Prussian blue-type cathode materials. Iron hexacyanoferrate (FeHCF) is considered a desirable Prussian blue-type cathode, but the incomplete electrochemical property of its low-spin iron sites hinders its further practical application. In this paper, carboxymethyl cellulose is demonstrated to have an appropriate binding energy through DFT calculations, synthesize Prussian blue in situ, balance Fe and water in FeHCF, and introduce Fe vacancies to activate low-spin Fe sites.
