Neutrophil isolation from nonhuman species.

Advances in the understanding of neutrophil biochemistry require the development of effective procedures for isolating purified neutrophil populations. Although methods for human neutrophil isolation are now standard, similar procedures for isolating neutrophils from many of the nonhuman species used to model human diseases are not as well developed. Because neutrophils are reactive cells, the method of isolation is extremely important to avoid isolation technique-induced alterations in cell function. We present methods here for reproducibly isolating highly-purified neutrophils from large (cow, horse, sheep) and small (mouse, rabbit) animal models and describe optimized details for obtaining the highest cell purity, yield, and viability. We also describe methods to verify phagocytic capacity in the purified cell populations using a flow cytometry-based phagocytosis assay.

Background: Cerebral blood flow is decreased in mouse models and patients of Alzheimer's disease (AD). We identified that about 2% of cortical capillaries in the APP/PS1 mouse model of AD had stalled blood flow due to neutrophils obstructing capillaries and contributing to vascular inflammation. Neutrophils are more reactive in AD.

Background: One of the major outstanding questions in the field of Alzheimer's disease (AD) research is the underlying mechanism by which APOE ε4, the strongest genetic risk factor for AD, contributes to disease pathogenesis. Current therapies targeting amyloid-beta plaques show modest effect in non-APOE4 male AD patients, and greatly increase the risk for amyloid-related imaging abnormalities - edema/effusion (ARIA-E) in APOE ε4 carriers. We made an important discovery that APOE4 neutrophil-microglia interactions drive cognitive impairment in a sex-dependent manner.
