The effect of tienamycin on selected parameters of the immunological system in mice infected with the influenza virus and Staphylococcus aureus was studied. Tienamycin was administered 24 hours, 2 hours prior the infection and 2 hours after the infection (model I), 24 hours and 48 hours after infecting the mice (model II). The authors analysed phagocytosis, bactericidal activity of peritoneal exudate cells and the LIF release induced by the staphylococcus antigen and the viral antigen by the spleen mice leukocytes. The authors found that the antibiotic given to non-infected mice inhibited phagocytosis of staphylococci by the peritoneal exudate cells. In mice that were infected a positive effect of the antibiotic on all studied parameters. Administering the antibiotic before infecting the mice produced an even better effect.

To determine the epidemiological, clinical and microbiological characteristics, of patients with post-traumatic osteomyelitis of extremity fractures, and provide evidence-based guidelines for early diagnosis and treatment, including empiric antibiotic therapy. Human subject research was performed using institutional review board approved protocols. A retrospective chart review was conducted on 5,368 patients diagnosed with extremity traumatic fractures from January 1, 2012 to December 31, 2015, to identify osteomyelitis patients.

Article Synopsis
  • Carbapenems are potent β-lactam antibiotics with a unique ability to resist inactivation by enzymes, and their biosynthesis involves distinct biochemical processes for complex and simple variants.
  • The carbapenem synthase enzyme, responsible for crucial final steps in producing simple carbapenems, can process two different substrate forms to create the same active antibiotic.
  • Research involving mutagenesis and structural analysis suggests a two-step reaction mechanism rather than a single cycle, involving more complexity in how the enzyme interacts with substrates during the antibiotic generation process.

ThnT is a pantetheine hydrolase from the DmpA/OAT superfamily involved in the biosynthesis of the β-lactam antibiotic thienamycin. We performed a structural and mechanistic investigation into the cis-autoproteolytic activation of ThnT, a process that has not previously been subject to analysis within this superfamily of enzymes. Removal of the γ-methyl of the threonine nucleophile resulted in a rate deceleration that we attribute to a reduction in the population of the reactive rotamer.

Article Synopsis
  • Carbapenems are a crucial class of antibiotics, with over 50 types that differ mainly by C-2 and C-6 side chains, affecting their effectiveness.
  • Researchers found that the ThnG and ThnQ enzymes from the thienamycin gene cluster in Streptomyces cattleya play key roles in oxidizing these side chains, enhancing the antibiotic properties of carbapenems.
  • Understanding how ThnG and ThnQ work can guide future studies on how carbapenem antibiotics are made, potentially leading to more effective treatments.

L-glutamate semialdehyde (L-GSA) is an intermediate in biosynthetic pathways including those leading to the carbapenem antibiotics. We describe studies on asymmetric deuteration or hydrogenation of appropriate didehydro-amino acid precursors for the stereoselective synthesis of C-2- and/or C-3-[2H]-labelled L-GSA suitable for use in mechanistic studies. Regioselective deuterium incorporation into the 5-position of L-GSA was achieved using a labelled form of the Schwartz reagent (Cp2Zr2HCl).
