HPLC/MS is now an essential method for the analysis of glycans and glycopeptides. A technique using ICATs is also becoming popular for their comparative/quantitative analysis based on MS signal intensities. However, the RP HPLC most often used causes "doublet" peaks of deuterium-labeled and nonlabeled peptides, which are well known for causing the chromatographic deuterium isotope effect. Such doublet peaks are undesirable for precise comparison of MS signal intensities. This report shows that, in the zwitterionic type of hydrophilic interaction chromatography, the chromatographic deuterium isotope effect is negligibly small for N-glycans derivatized with deuterium-labeled 2-aminopyridine and N-glycopeptides derivatized with deuterium-labeled succinic anhydride.

A new approach for hydrogen isotope separation using an unsaturated organometallic complex was proposed. Adsorption measurements of [Mn(dppe)(CO)(N)](BArF) (Mn-dppe) (dppe = 1,2-bis(diphenylphosphino)ethane, BArF = B[CH(3,5-CF)]) using H and D revealed a significant difference in the adsorption enthalpy of H/D at much higher room temperatures than in previous studies, with D molecules being more strongly adsorbed on unsaturated metal sites. Mixed gas adsorption isotherms were calculated at each temperature using IAST, and it was predicted that D uptake was much larger than H uptake.

Background: Pegfilgrastim-cbqv/CHS-1701 (UDENYCA) (hereafter referred to as pegfilgrastim-cbqv) was approved in 2018 by the US Food and Drug Administration as a biosimilar for pegfilgrastim (Neulasta) (hereafter referred to as pegfilgrastim). Both pegfilgrastim-cbqv and pegfilgrastim are conjugates of recombinant human granulocyte colony stimulating factor (r-metHuG-CSF) with a 20 kDa polyethylene glycol (PEG) indicated to decrease the incidence of infection, as manifested by febrile neutropenia, in patients receiving myelosuppressive anticancer drugs. The demonstration of analytical similarity for PEG-protein conjugates presents unique challenges since both the protein and PEG attributes must be characterized.

Hydrogen/deuterium (H/D) isotope effects are not unusual in chromatography and such phenomena have been observed in both gas- and liquid-phase separations. Despite the numerous reports on this topic, the understanding of mechanisms and the underlying noncovalent interactions at play remains rather challenging. In our recent study, we reported baseline separation of isotopologoues of some amphetamine (AMP) derivatives on achiral and polysaccharide-based chiral columns, as well as some correlations between the degree of separation of enantiomers and isotopologues on (the same) polysaccharide-based chiral column(s).
