Kinetic, thermodynamic, and ab initio insights of AsnGly isomerisation as a ticking time bomb for protein integrity.

Commun Chem

December 2024

Laboratory of Structural Chemistry and Biology, Institute of Chemistry, ELTE Eötvös Loránd University, Pázmány Péter sétány 1/A, 1117, Budapest, Hungary.

Under physiological conditions in peptides or proteins, the -AsnGly- motif autonomously rearranges within hours/days to β-Asp and α-Asp containing sequence, via succinimide intermedier. The formation of the succinimide is the rate-limiting step, with a strong pH and temperature dependence. We found that Arg(+) at the (n + 2) position (relative to Asn in the n position) favors isomerisation by forming a transition-state like structure, whereas Glu(-) disfavors isomerisation by adopting a β-turn like conformer.

CircMIB1 inhibits glioma development and progression through a competing endogenous RNA interaction network.

Front Mol Biosci

December 2024

Department of Emergency Medicine and Laboratory of Emergency Medicine, West China Hospital, West China School of Medicine, Sichuan University Chengdu, Chengdu, China.

Introduction: The critical role of circular RNAs as non-coding RNAs in glioma has been extensively investigated. Therefore, we aimed to explore the role and potential molecular mechanisms of circRNA-mind bomb homolog 1 (circMIB1) in gliomas.

Methods: RNA sequencing was used to analyze the expression profiles of circRNAs in glioma tissues and normal brain tissues.

Behcet's disease is a rare, chronic inflammatory disorder that can affect various organs, with large vessel involvement being particularly uncommon. This case report discusses a 17-year-old male with Behcet's disease who presented with a rapidly enlarging, painful mass in the right groin region, later diagnosed as a femoral artery aneurysm. The patient underwent successful surgical intervention and was subsequently managed with immunosuppressive therapy to prevent relapse.

Adolescent and young adult (AYA, 15-39 years old) patients with papillary thyroid carcinoma (PTC) experience significant psychological distress and place great importance on obtaining information regarding the disease; however, their demands remain unmet. We aimed to investigate clinicopathological and molecular features of PTC in AYA patients and compare them to those of PTC in older patients (≥40 years). This retrospective study enrolled 1,677 patients diagnosed with PTC from January 2018 to December 2022, with 400 AYA patients and 1,277 older adults.

Article Synopsis
  • Carbon reserves in trees are influenced by past photosynthesis and can be dated using radiocarbon dating, allowing researchers to study their distribution and turnover.
  • A new process model was created to analyze how different tree species manage their carbon reserves, revealing that both young and old reserves coexist within a single tree and are significantly mixed.
  • Disturbances like drought, fire, and pests negatively affect reserve mixing and longevity, with older reserves showing notable age shifts after disturbances, suggesting that ecological variations across species play a critical role in reserve management.
