This unit describes procedures for generating long-range restriction maps of genomic DNA and for analysis of large insert clones. The basic protocol details restriction digestion of agarose-embedded DNA, PFGE separation, Southern transfer, and hybridization. Support protocols describe the preparation of high-molecular-weight genomic DNA samples in agarose blocks and in agarose microbeads, respectively. Additional support protocols describe the preparation of DNA size standards from l phage and two yeast species, Saccharomyces cerevisiae and Schizosaccharomyces pombe. An alternative method of preparing S. cerevisiae size standards using lithium dodecyl sulfate (LiDS) solubilization is provided. The final protocol details the preparation of BAC DNA suitable for digestion, mapping, and sequencing.

Expanding toolkits of EPA/DHA enrichment from natural sources is essential for better satisfying increasing demands for them. Lipase K80, from Proteus vulgaris K80, showed an application potential in EPA/DHA enrichment, whereas no desired heterologous expression in generally regarded as safe (GRAS) hosts restricted its relevant applications. In this study, expression of lipase K80 in a well-reputed GRAS host, Pichia pastoris, was achieved and further enhanced via combining disruption of its C-terminal KKL motif with co-expression of N-Acetyltransferase Mpr1, with a cumulative increment of nearly 200 % in the secretion level and the volumetric activity.

The crystallographic restriction theorem constrains two-dimensional nematicity to display either Ising (Z_{2}) or three-state-Potts (Z_{3}) critical behaviors, both of which are dominated by amplitude fluctuations. Here, we use group theory and microscopic modeling to show that this constraint is circumvented in a 30°-twisted hexagonal bilayer due to its emergent quasicrystalline symmetries. We find a critical phase dominated by phase fluctuations of a Z_{6} nematic order parameter and bounded by two Berezinskii-Kosterlitz-Thouless (BKT) transitions, which displays only quasi-long-range nematic order.

Selection and dispersal are the primary processes influencing community assembly at both global and regional scales. Although the effectiveness of dispersal is often examined within the same biome, microscopic organisms demonstrate the capability to colonise and thrive across different biomes. In this study, we evaluated the relationship between (i) aquatic, (ii) sedimentary and (iii) aerial microbial communities, and how local selective pressures influence the potential impact of inter-biome dispersal, focusing on the salinity gradient stress over time in ephemeral saline lakes.
