Melanocortins and their implication in melanoma.

The melanocortins (MCs), that is, the melanocyte-stimulating hormones (MSHs) and ACTH, are a group of related peptides containing the typical melanotropin core sequence, His-Phe-Arg-Trp, and are derived from a common precursor, pro-opiomelanocortin. They are pleiotropic molecules that occur in the pituitary, some brain regions, and also in several peripheral tissues, and they exert a variety of physiologic functions. Their effect on melanogenesis in the skin is well established, but their role in melanocyte and melanoma cell proliferation and metastasis is less clear. The recent cloning of five types of MC receptors (MC1-5), new studies on the regulation of these receptors, the discovery of a naturally occurring MSH antagonist, the agouti protein, and the finding that melanocytes and melanoma cells exclusively express MC1 receptors have laid the basis for the future development of specific MC ligands, which may become useful for melanoma diagnosis and eventually therapy.

N-Branched Tricyclic Guanidines as Novel Melanocortin-3 Receptor Agonists and Melanocortin-4 Receptor Antagonists.

The melanocortin receptors are a class of centrally and peripherally expressed G protein-coupled receptors, of which the MC3R and MC4R subtypes are implicated in the regulation of appetite and energy homeostasis and can serve as potential therapeutic targets for disorders such as obesity and cachexia. An unbiased high-throughput mixture-based library screen was implemented to identify novel ligands with an emphasis on the identification of nanomolar-potent agonists of the mouse melanocortin-3 receptor. This screen yielded the discovery of an N-branched tricyclic guanidine scaffold (TPI2408) that contained three nanomolar potent mMC3R agonists and additional compounds that possessed antagonism for the mMC4R.

Aim: To determine the relationship between alpha Melanocyte Stimulating Hormone (α-MSH) and Melanocortin-4 receptor (MCR4) levels and sexual function levels of patients with and without SSRI related SD and control group and to examine whether α-MSH and MCR4 play a role in the etiology of SSRI related SD.

Methods: A total of 92 patients and 49 healthy volunteers who applied to psychiatry outpatient clinic were included in the study.

In parallel, normotensive adrenal glands frequently harbor aldosterone-producing micronodules with pathogenic somatic mutations known to induce primary aldosteronism (PA). A deeper understanding of these phenomena would inform the origins of PA and its role in hypertension pathogenesis.
