The reaction of phosphonium alkylidenes [(H2IMes)RuCl2=CHPR3]+[A]- (R = C6H11, A = OTf or B(C6F5)4, 1-Cy; R = i-C3H7, A = ClB(C6F5)3 or OTf, 1-iPr) with 1 equiv of ethylene at -78 degrees C, in the presence of 2-3 equiv of a trapping olefin substrate, yields intermediates relevant to olefin metathesis catalytic cycles. Dimethyl cyclopent-3-ene-1,1-dicarboxylate gives solutions of a substituted ruthenacyclobutane 3 of relevance to ring closing metathesis catalysis. 1H and 13C NMR data are fully consistent with its assignment as a ruthenacyclobutane, but 1JCC values of 23 Hz for the CalphaH2-Cbeta bond and 8.5 Hz for the CalphaH-Cbeta bond point to an unsymmetrical structure in which the latter bond is more activated than the former. In contrast, trapping with acenaphthylene leads to an olefin carbene complex (6) in which the putative ruthenacyclobutane has opened; this species was also fully characterized by NMR spectroscopy and compared to related species reported previously.

Efficient methods for synthesizing allenes from readily available starting materials pose a persistent challenge in organic chemistry. In this work, we present a novel two-stage protocol for allene synthesis involving the single-atom insertion into alkenes, facilitated by synergistic photoredox and cobalt catalysis. Diverging from conventional methods such as the Doering-LaFlamme reaction, this photochemical rearrangement approach operates efficiently under mild conditions in a radical-based manner.

A pronounced nucleophilicity in combination with a distinct redox non-innocence is a unique feature of a coordinated ligand, which in the current case, leads to unprecedented carbon-centered reactivity patterns: A carbodiphosphorane-based (CDP) pincer-type rhodium complex allows to cleave two C-Cl-bonds of geminal dichlorides via two consecutive SN2-type oxidative additions resulting in the formation of a stabilized carbene fragment. In the presence of a suitable reductant the carbene fragment can even be converted into olefines or hydrodehalogenation products in a catalytic reaction. The developed method can also be used to convert chlorofluorocarbons (CFCs) such as CH2ClF to fluoromethane and methane.

Machine-Learning-Aided Engineering Hemoglobin as Carbene Transferase for Catalyzing Enantioselective Olefin Cyclopropanation.

JACS Au

Key Laboratory of Molecular Enzymology and Engineering of Ministry of Education, School of Life Sciences, Jilin University, Changchun 130023, P. R. China.

In this study, we developed a machine-learning-aided protein design strategy for engineering hemoglobin (VHb) as carbene transferase. A Natural Language Processing (NLP) model was used for the first time to construct an algorithm (EESP, enzyme enantioselectivity score predictor) and predict the enantioselectivity of VHb. We identified critical amino acid residue sites by molecular docking and established a simplified mutation library by site-saturated mutagenesis.

Visible-Light-Mediated Radical -Hydroboration of Alkynes with NHC Borane.

School of Pharmaceutical Science, Hengyang Medical School, University of South China, Hengyang 421001, Hunan, China.

Although the radical hydroboration of alkenes with N-heterocyclic carbene (NHC) borane is well documented, the radical hydroboration of alkynes, especially terminal alkynes, remains challenging. Herein, a photoredox-catalyzed radical -hydroboration of alkynes with NHC borane has been developed, which provided various alkenyl boron compounds in moderate to good yields. This protocol exhibits a broad substrate scope, as both internal and terminal alkynes were compatible.

Sulfenyl fluorides are organic compounds of sulfur in formal oxidation state +2 with the formula R-S-F. Although the chloride, bromide, and iodide analogues have been extensively described in the literature, arenesulfenyl fluorides remain essentially unstudied. These structures have been implicated as putative intermediates in established processes to access polyfluorinated sulfur species; however, definitive and direct evidence of their existence has not been obtained, nor has a systematic understanding of their reactivity.
