Epigenetic mechanisms involving dynamic changes in posttranslational histone modifications commonly control gene transcription and therefore the execution of all cellular differentiation programs. The differentiation of hematopoietic stem cells into specific progenitor cells and the diverse blood cell types represents a particularly powerful system for the study of epigenetic mechanisms. The hematopoietic system allows one to define mechanisms underlying the establishment and regulation of histone modification patterns covering entire genes and/or chromosomes at distinct stages of differentiation. This chapter reviews progress in elucidating principles underlying epigenetic control of complex loci, specifically focusing on genes differentially expressed during hematopoiesis.

J-Lat cells are derivatives of the Jurkat CD4 T cell line that contain a non-infectious, inducible HIV provirus with a GFP tag. While these cells have substantially advanced our understanding of HIV latency, their use by many laboratories in low and middle-income countries is restricted by limited access to flow cytometry. Upon infection, human papillomavirus (HPV) manipulates host cell gene expression to create an environment that is supportive of a productive and persistent infection. Its action on sperm is of particular interest, since they transfer genetic and epigenetic information to the offspring, including microRNAs, configuring a mechanism of paternal epigenetic inheritance. MicroRNAs are known to participate in a wide variety of mechanisms in basically all cells and tissues, including the brain and the sperm cells, which are known, respectively, to present 70% of all identified microRNAs and to transfer these molecules to the embryo.
