The Hit-to-Lead-to-Candidate process continues to evolve rapidly, and while technological advances offer much potential, the reality often pales to the promise. Conversely, strategies and tactics implementing existing technologies may result in more benefit in the end. This article focuses on some of the thinking and approaches that may improve the efficiency and effectiveness of the beginnings of the drug discovery path. From the perspective of computational chemists, different types of strategy and philosophy of approach will be treated including: considerations of early lead choices, strategies for improving poor leads, multivariate optimization, opportunities for informatics, and engineering good decisions.

Intimate partner violence (IPV) is a significant public health issue affecting many women worldwide. While extensive research exists on IPV during pregnancy and postpartum, there is limited information on IPV against mothers during the critical child-rearing stage, specifically the first three years following childbirth. This study examines the prevalence and patterns of IPV among mothers in China during this stage, identifying associated factors across four family subsystems: individual, husband-and-wife, mother-child, and family context, to guide the development of tailored prevention strategies.

There is an urgent necessity to devise efficient tactics to tackle the inevitable development of resistance to osimertinib, which is a third-generation epidermal growth factor receptor (EGFR) inhibitor used in treating EGFR-mutant nonsmall cell lung cancer (NSCLC). This study demonstrates that combining itraconazole with osimertinib synergistically reduces the proliferation and migration, enhances the apoptosis of osimertinib-resistant cells, and effectively inhibits the growth of osimertinib-resistant tumors. Mechanistically, itraconazole combined with osimertinib promotes the proteasomal degradation of sonic hedgehog (SHH), resulting in inactivation of the SHH/Dual-specificity phosphatase 13B (DUSP13B)/p-STAT3 and Hedgehog pathways, suppressing Myc proto-oncogene protein (c-Myc).

Parasitoid wasps exert strong selective pressure on their hosts, driving the evolution of diverse defense strategies. Drosophila, a widely studied model organism, hosts a wide range of parasites, including parasitoid wasps, and has evolved immune and behavioral mechanisms to mitigate the risk of parasitization. Integrated Pest Management (IPM) provides a powerful framework for addressing threats to human well-being caused by nuisance species including invasives. We examined the hypothesis that adaptive management could erode barriers to IPM implementation by developing a decision-analytic adaptive management framework for invasive sea lamprey (Petromyzon marinus) IPM in the Laurentian Great Lakes of North America. The framework addressed objectives associated with coordinating multiple sea lamprey control actions at the regional scale and objectives associated with internal validity of control actions.
