Pathogenicity of Trypanosoma brucei brucei in experimentally infected pigs.

Department of Veterinary Medicine, University of Ibadan, Nigeria.

Published: October 1992

An experimental infection of 4-to 5-month old pigs with a stock of Trypanosoma brucei brucei resulted in a high parasitaemia, anorexia, pyrexia and a decline in the packed cell volume by one third. Nervous sign of circling and wobbling of the hind legs occurred in one of the pigs which at necropsy revealed a very severe meningo-encephalitis and the presence of trypanosomes in the brain. These results confirm that T. b. brucei might cause a severe disease in pigs.

To regain infectivity, Trypanosoma brucei, the pathogen causing Human and Animal African trypanosomiasis, undergoes a complex developmental program within the tsetse fly known as metacyclogenesis. RNA-binding protein 6 (RBP6) is a potent orchestrator of this process, however, an understanding of its functionally important domains and their mutational constraints is lacking. Here, we perform deep mutational scanning of the entire RBP6 primary structure.

RNA interference in protozoan parasites and its application.

J Microbiol Immunol Infect

January 2025

Department of Urology, Tungs' Taichung Metro Harbor Hospital, Taichung, Taiwan. Electronic address:

RNA interference (RNAi) is a biological process in which RNA molecules are involved in sequence-specific suppression of gene expression, via small RNA triggers derived from double-stranded RNA that can target specific genes; it is a natural process that plays a role in both the regulation of protein synthesis and in immunity. Discovery of RNAi by Fire and Mello in 1998 had a profound impact on unraveling novel aspects of eukaryotic biology. RNA interference (RNAi) has proven to be an immensely useful tool for studying gene function and validation of potential drug targets in almost all organisms.

Antileishmanial and Antitrypanosomal Trends of Synthetic Tetralone Derivatives.

Drug Dev Res

February 2025

Department of Pharmaceutical Chemistry, College of Pharmacy, Jouf University, Sakaka, Aljouf, Saudi Arabia.

Leishmaniasis and trypanosomiasis are parasitic diseases that are closely linked to poverty, pose significant local burdens, and are common in tropical and subtropical regions. Various synthetic tetralone derivatives were studied as potential scaffolds for antileishmanial and antitrypanosomal activities. The compounds were studied for their effectiveness against multiple kinetoplastid protozoan pathogens: Leishmania major, Leishmania mexicana, and bloodstream trypomastigotes of Trypanosoma brucei brucei.

Canine African trypanosomosis is endemic in sub-Saharan Africa. Chemotherapy remains the commonly employed approach to trypanosomosis control. However, it is beleaguered by the absence of new drugs, treatment failures, relapse infection and resistance.

Human African trypanosomiasis (HAT) is one of the most lethal of the neglected tropical diseases. While the discovery of a novel antitrypanosomal drug is highly desired, the creation of a superior lead compound is challenging. Herein we report ukabamide (), which was isolated from a marine sp.
