Chromatography is the most widely used technique for the purification of biopharmaceuticals in the biotech industry. Surprisingly, process development is often still based on empirical studies or experience; recently high-throughput screening stations are employed to minimize development time and to improve screening quality. Still, experimental effort remains high and a more detailed understanding of adsorption mechanisms on a molecular level underlying chromatographic separation could help in the future to select and design chromatography steps in silico. In this study, we focused on the elucidation of protein orientation upon adsorption onto a chromatographic resin. We identified two characteristic binding sites of lysozyme on SP Sepharose Fast Flow and one multipoint interaction of lysozyme with SP Sepharose XL. Increasing ionic strength did not significantly influence the binding, whereas changes in the mobile phase pH led to a re-orientation on SP Sepharose FF. This phenomenon agrees well with theoretical considerations, including a detailed description of the surface charge distribution with changing pH and linear elution experiments, giving an idea why proteins are often retained on ion-exchange materials beyond their isoelectric point.

The one-dimensional nanomaterials known as nanofibers have remarkable qualities, such as large surface areas, adjustable porosity, and superior mechanical strength. Ionomers, types of polymers, have ionic functional groups that give them special properties, including high mechanical strength, water absorption capacity, and ionic conductivity. Integrating ionomers and nanofibers with diverse materials and advanced methodologies has been shown to improve the mechanical strength, processing capacity, and multifunctional attributes of ionomeric nanofibers.

In this article, we report on the alginate heterografted by Poly(N-isopropyl acrylamide-co-N-tert-butyl acrylamide) and Poly(N-isopropyl acrylamide) (ALG-g-P(NIPAM86-co-NtBAM14)-g-PNIPAM) copolymer thermoresponsive hydrogel, reinforced by substituting part of the 5 wt% aqueous formulation by small amounts of Poly(acrylic acid)-g-P(boc-L-Lysine) (PAA-g-P(b-LL)) graft copolymer (up to 1 wt%). The resulting complex hydrogels were explored by oscillatory and steady-state shear rheology. The thermoresponsive profile of the formulations were affected remarkably by increasing the PAA-g-P(b-LL) component of the polymer blend.

Water pollution is a major global issue, and antibiotic drugs released into aquatic environments by the pharmaceutical industry, such as ciprofloxacin, have negative consequences on both human health and the ecosystem. In this study, the performance of PVA as a polymer ligand for ciprofloxacin (CPFX) removal is evaluated through polymer-enhanced ultrafiltration using a novel composite PVC-ZnO membrane. The initial concentration of the ciprofloxacin solution, pH, ionic strength, ideal polymer concentration, duration, and maximum retention capacity were among the factors that were examined.

Fibrotic cardiomyopathy represents a significant pathological condition characterized by the interaction between cardiomyocytes and fibroblasts in the heart, and it currently lacks an effective cure. In vitro platforms, such as engineered heart tissue (EHT) developed through the co-culturing of cardiomyocytes and fibroblasts, are under investigation to elucidate and manipulate these cellular interactions. We present the first integration of mathematical electrophysiological models that encapsulate fibroblast-cardiomyocyte interactions with experimental EHT studies to identify and modulate the ion channels governing these dynamics.
