This unit describes methods for passaging and harvesting the basement membrane matrix-producing EHS tumor and for the subsequent isolation of a crude mixture of basement membrane components termed Matrigel, which promotes the differentiation of a variety of epithelial, endothelial, and neuronal cells. Procedures for the isolation of the adhesive glycoprotein laminin-1 and of type IV collagen are also included. Support protocols cover the maintenance and harvesting of EHS tumors in mice and maintenance of mice on a lathrogenic diet.

Butterfly wing eyespots are developmentally determined at the early pupal stage, when prospective eyespot focal cells underneath the pupal cuticle focal spot function as eyespot organizers in the pupal wing tissue. Here, we performed light microscopy and transmission electron microscopy (TEM) to describe cellular structures of pupal wing tissue with an eyespot organizer immediately after pupation using the Blue Pansy butterfly Junonia orithya. The pupal forewing dorsal epidermis was a pseudostratified monolayer of vertically elongated epidermal cells.

Background: APOEε4 significantly increases the risk of developing Alzheimer's disease (AD). Cognitively healthy APOEε4-carriers exist, suggesting potential protective mechanisms against APOEε4. We hypothesized that some APOEε4-carriers may have genetic variations protecting them from developing APOEε4-mediated AD pathology.

Background: Mechanisms driving cerebrovascular decline during Alzheimer's disease and related dementias (ADRD) are poorly understood. Methylenetetrahydrofolate reductase (MTHFR) is an enzyme in the folate/methionine pathway. Variants in MTHFR, notably 677C>T, are associated with ADRD.

Background: Both intrinsic renal cells and immune cells contribute to driving renal inflammation and damage. However, the respective roles of intrinsic renal cells and immune cells in crescentic glomerulonephritis, and the key molecular factors driving pathogenesis are still unclear.
Methods: The roles of intrinsic renal cells and renal infiltrating immune cells in crescent formation were explored using renal transplantation after experimental anti-GBM disease induction in 129x1/svJ and C57BL/6J mice.
