Background & Aims: Glucocorticoids, produced by the adrenal gland under control of the hypothalamic-pituitary-adrenal axis, exert their metabolic actions largely via activation of the glucocorticoid receptor (GR). Synthetic glucocorticoids are widely used as anti-inflammatory and immunosuppressive drugs but their application is hampered by adverse metabolic effects. Recently, it has been shown that GR may regulate several genes involved in murine bile acid (BA) and cholesterol metabolism, yet the physiological relevance hereof is controversial.

The influence of calcitonin on the choleresis level and bile acids spectrum was investigated in acute experiments on the rats with common biliary duct cannulated. We determined the intensity of bile secretion during 3 hours and quantitative content of conjugated and free bile acids using thin-layer chromatography. It was shown that intramuscular introduction of synthetic salmon calcitonin (200 ng/kg) increases the secreted bile volume and the content of conjugated bile acids in the bile.

One of the synthetic analogues of antidiuretic hormone-desmopressin is used in patients with central diabetes insipidus and in those with coagulation disorders. However, its effects on bile secretion are not fully defined. We investigated the effect of desmopressin on bile formation and determined the role of V1a vasopressin receptors in the action of desmopressin on choleresis.

It is shown that desmopressin increases bile flow and concentration of taurin-conjugated bile acids. In this situation, the concentration ofglycocholates practically is not changed, and concentration of free bile acids is diminished, which results in increase in the conjugation coefficient.

New analogs of cyclic amino acid-conjugated bile acids were synthesized, and their physicochemical and biological properties were compared with those of natural analogs. Ursodeoxycholic acid was amidated with D-proline, L-proline, 4-hydroxy-L-proline, and 4-methoxy-L-proline. Hyocholic and hyodeoxycholic acids were amidated with L-proline.
