Tomato is an economically important crop and a significant dietary source of important phytochemicals, such as carotenoids and flavonoids. Although it has been known for many years that the plant hormone ethylene is essential for the ripening of climacteric fruits, its role in fruit growth and maturation is much less well understood. In this study, data are presented which indicate that fruit-specific suppression of the ethylene receptor LeETR4 causes early ripening, whereas fruit size, yield and flavour-related chemical composition are largely unchanged. Early fruit ripening is a highly desirable and valuable trait, and the approach demonstrated here should be applicable to any fruit species requiring ethylene to ripen. These results demonstrate that ethylene receptors probably act as biological clocks regulating the onset of tomato fruit ripening.

Flavonoids have a major contribution to the fruit quality in cultivated strawberries and are regulated by MYB, bHLH and WD40 transcriptional factors. We reported here the identification of the FaMYB5, an R2R3-MYB transcription factor, which positively regulated the accumulation of anthocyanins and proanthocyanidins through the trans-activation of the F3'H and LAR. The strawberry FaEGL3 and FaLWD1/FaLWD1-like interact with the R2R3-FaMYB5 to form an MYB-bHLH-WD40 complex (MBW), enhancing the regulatory efficiency.

Spontaneous mutations in fruit-specific carotenoid biosynthetic genes of tomato (Solanum lycopersicum) have led to improved understanding of ripening-associated carotenogenesis. Here, we confirm that ZDS is encoded by a single gene in tomato transcriptionally regulated by ripening transcription factors RIN, NOR and ethylene. Manipulation of ZDS was achieved through transgenic repression and heterologous over-expression in tomato.

Phosphoenolpyruvate carboxykinase (PEPCK) is a key regulatory enzyme and is utilized in the gluconeogenesis pathway in plants. Although, its catalytic and regulatory properties are quite well understood, there are uncertainties regarding its physiological role in many plants tissues such as the flesh of developing fruits. To further understand the function of PEPCK in fruits and other tissues, RNAi transgenic tomato plants in which SlPEPCK transcription was down-regulated by either CaMV 35S constitutive promoter or the fruit-specific E8 promoter were generated and characterized on the basis of their phenotypic and metabolic aspects.
