The effectiveness of an in vivo synthesis of antibodies to clenbuterol with the use of gold nanoparticles as a carrier was evaluated. For comparison, conjugates of clenbuterol with bovine serum albumin were used in immunization. The serum titer was determined by an ELISA. The antibodies were tested by an immunodot assay with immunogold markers. With both techniques we obtained specific and relatively high-titer antibodies to clenbuterol. It was found that the antibodies obtained with the use of gold nanoparticles were not inferior in titer to those obtained to the conjugates of clenbuterol with the protein but surpassed them in specificity.

A highly sensitive lateral flow immunoassay (LFIA) is developed for the enzyme-catalyzed and double-reading determination of clenbuterol (CLE), in which a new type of probe was adopted through the direct electrostatic adsorption of ultra-small copper-gold bimetallic enzyme mimics (USCGs) and monoclonal antibodies. In the assay, based on the peroxidase activity of USCG, the chromogenic substrate TMB-HO was introduced to trigger its color development, and the results were compared with those before catalysis. Bimodal-type multiplexed immunoassays with complementary mode-based correlation analysis are gaining increasing attention for enhancing the practicability of the lateral flow immunoassay (LFIA). Nonetheless, the restriction in visually indistinguishable multitargets induced by a single fluorescent color and difficulty in single acceptor ineffectual fluorescence quenching due to the various spectra of multiple different donors impede the further execution of colorimetric-fluorescence bimodal-type multiplexed LFIAs. Herein, the precise spectral overlap-based donor-acceptor pair construction strategy is proposed by regulating the size of the nanocore, coating it with an appropriate nanoshell, and selecting a suitable fluorescence donor with distinct colors.

A wide variety of small-molecule pollutants is harmful to human health, and their highly sensitive universal and rapid detection in complex environments remains a challenge. Herein, a multiplexed and ultrasensitive immunochromatographic strip (ICS) was developed for the universal analysis of three kinds of different pollutants based on multilayered fluorescent nanofilm-guided signal amplification. Flexible three-dimensional nanofilms (GO-MQD) with large surface areas, high quantum dot (QD) loading, superior luminescence, and good stability were synthesized through the electrostatic adsorption-mediated layer-by-layer assembly of three layers of small QDs onto two-dimensional graphene oxide (GO) nanosheets, modified with specific antibodies, and utilized as enhanced fluorescent tags in the ICS method for quantitative target detection.
