Loss of H4 lysine 16 (H4K16) acetylation was shown to be a common feature in human cancer. However, it remained unclear which enzyme is responsible for the loss of this modification. Having recently identified the histone acetyltransferase human MOF (hMOF) to be required for bulk H4K16 acetylation, here we examined the involvement of hMOF expression and H4K16 acetylation in breast cancer and medulloblastoma. Analysis of a recent mRNA expression profiling study in breast cancer (n = 100 cases) and an array-CGH screen in medulloblastomas (n = 102 cases), revealed downregulation in 40% and genomic loss in 11% of cases, respectively. We investigated hMOF protein expression as well as H4K16 acetylation in large series of primary breast carcinomas (n = 298) and primary medulloblastomas (n = 180) by immunohistochemistry. In contrast to nontransformed control tissues, significant fractions of both primary breast carcinomas and medulloblastomas showed markedly reduced hMOF mRNA and protein expression. In addition, hMOF protein expression tightly correlated with acetylation of H4K16 in all tested samples. For medulloblastoma, downregulation of hMOF protein expression was associated with lower survival rates identifying hMOF as an independent prognostic marker for clinical outcome in univariate as well as multivariate analyses.

The oomycete Phytophthora infestans is one of the most destructive phytopathogens globally. It has a proven ability to adapt to changing environments rapidly; however, molecular mechanisms responsible for host invasion and adaptation to new environmental conditions still need to be explored. The study aims to understand the epigenetic mechanisms exploited by P.

Article Synopsis
  • Autosomal-dominant inheritance is a key factor in familial Alzheimer's disease (FAD), but it explains only a small part of its cases; thus, the study explored epigenetic memory as a potential contributor to FAD development across generations.
  • Researchers found that a specific gene, mys-2, which affects histone modification (H4K16 acetylation), influenced the occurrence of Alzheimer's disease, with beneficial changes in parent generation being passed down to offspring through epigenetic memory.
  • The study suggests that the MYS-2/MOF protein could be inherited epigenetically, highlighting a novel perspective on the causes and transmission of FAD.

Background: As the leading cause of end-stage liver disease, nonalcoholic fatty liver disease (NAFLD) is mainly induced by lipid dyshomeostasis. The translation of endogenous circular RNAs (circRNAs) is closely related to the progression of various diseases, but the involvement of circRNAs in NAFLD has not been determined.

Methods: Combined high-throughput circRNA profiles were used to identify circRNAs with translational potential.

Article Synopsis
  • - The study highlights that abnormal trophoblast self-renewal and differentiation in early pregnancy can lead to miscarriages, with the KAT8 enzyme playing a critical role in regulating these processes.
  • - Researchers found that deleting KAT8 in trophoblasts causes severe developmental issues, but boosting the expression of a key marker (CDX2) can partially counteract these defects.
  • - Changes in the levels of KAT8, CDX2, and H4K16ac are linked to recurrent pregnancy loss, suggesting that targeting the KAT8-H4K16ac-CDX2 pathway might be a potential treatment option for this issue.

Lysine acetyltransferase 8, also known as KAT8, is an enzyme involved in epigenetic regulation, primarily recognized for its ability to modulate histone acetylation. This review presents an overview of KAT8, emphasizing its biological functions, which impact many cellular processes and range from chromatin remodeling to genetic and epigenetic regulation. In many model systems, KAT8's acetylation of histone H4 lysine 16 (H4K16) is critical for chromatin structure modification, which influences gene expression, cell proliferation, differentiation, and apoptosis.
