While users of web-based information often report their reliance on such information for medical decision making, it has yet to be determined if this is universally true across all types of information or specialty domains. Some have argued that the web may be improperly used as 'substituted clinical judgement', rather than serving as a support tool for patients and their doctors. Further, little attention has been paid to the selective development of methodologies for consumer-centred discussions, or to selective grouping and analysis of debated 'domains of uncertainty' in healthcare. Our objective in this study is to introduce a more refined qualitative model for discussion group or chatroom evaluation than has traditionally been used, and illustrate the application of grounded theory as an inductive framework, using assessment of e-discussion within an area of ongoing medical uncertainty, narcolepsy. Using this approach we find that while consumers often debate and discuss topics traditionally reserved for the doctor-patient relationship, they routinely encourage provider advice and dialogue within such discussions, even after experiencing unsatisfactory outcomes in such settings.

Human epidermal growth factor receptor 2 (HER2, also known as ERBB2) signaling promotes cell growth and differentiation, and is overexpressed in several tumor types, including breast, gastric and colorectal cancer. HER2-targeted therapies have shown clinical activity against these tumor types, resulting in regulatory approvals. However, the efficacy of HER2 therapies in tumors with HER2 mutations has not been widely investigated.

Dual inhibition of the angiopoietin (Ang)/Tie and vascular endothelial growth factor (VEGF) signalling pathways in patients with retinal diseases, such as neovascular age-related macular degeneration (nAMD) and diabetic macular oedema (DME), may induce greater vascular stability and contribute to increased treatment efficacy and durability compared with treatments that only target the VEGF pathway. Faricimab, a bispecific intravitreal agent that inhibits both VEGF and Ang-2, is the first injectable ophthalmic drug to achieve treatment intervals of up to 16 weeks in Phase 3 studies for nAMD and DME while exhibiting improvements in visual acuity and retinal thickness. Data from real-world studies have supported the safety, visual and anatomic benefits and durability of faricimab, even in patients who were previously treated with other intravitreal agents.

Peripheral nerve injury (PNI) as a common clinical issue that presents significant challenges for repair. Factors such as donor site morbidity from autologous transplantation, slow recovery of long-distance nerve damage, and deficiencies in local cytokines and extracellular matrix contribute to the complexity of effective PNI treatment. It is extremely urgent to develop functional nerve guidance conduits (NGCs) as substitutes for nerve autografts.

Rehabilitation is the process of helping people regain or improve lost or impaired function due to injury, illness, or disease. To assist in tracking the progress of patients undergoing rehabilitation, this paper proposes a lightweight graph-based deep-learning model for the automatic assessment of physical rehabilitation exercises. The model takes as input the 3D skeleton sequence of a patient performing a movement and outputs a continuous quality score, as a means for patient supervision that could complement or even substitute the need for ordinary clinical exams.
