Objective: We examined the transcriptional response to serum stimulation as an in vitro model of wound healing in keloid fibroblasts to identify molecular mechanisms leading to their aberrant growth.
Summary Background Data: Keloids are proliferative dermal growths representing a pathologic wound healing response. Although several groups have shown increased expression of profibrotic factors in keloids, there is little known about why they are expressed at higher levels than normal.
Methods: Fibroblasts derived from keloids and normal scar were subjected to serum stimulation as an in vitro model to mimic a component of the wound microenvironment to examine differential gene expression in keloid derived fibroblasts versus normal human fibroblasts. A promoter analysis was performed to identify specific enhancers involved in mediating the differential response of connective tissue growth factor (CTGF, CCN2). Point mutations in the enhancers were performed to confirm their role. Finally, we examined activation of transcription factors known to bind the targeted enhancers.
Results: Transcription of CCN2 after serum stimulation was significantly higher in keloid versus normal fibroblasts. Promoter analysis demonstrates the fragment from -625/-140 conferred increased serum responsiveness. Mutational analysis showed an AP-1 and SMAD binding site were both necessary for serum responsiveness. Preventing activation of either transcriptional complex will block CCN2 transcription. Additional experiments suggest that a single complex that includes components of the AP-1 and SMAD binding complexes is responsible for transactivation in response to serum. The key difference between keloid and normal fibroblasts appears to be the degree of activation of c-Jun.
Conclusions: We suggest that altered responsiveness to cellular stress, based upon current data using serum stimulation and past data on response to mechanical strain, is a key defect leading to keloid formation.

The objective was to determine the effects of injectable trace minerals (ITM, containing Se, Cu, Zn & Mn) administered at the time of primary intranasal (IN) modified-live virus (MLV) vaccination of young dairy calves on the serum neutralizing antibody (SNA) titers to Bovine herpes virus 1 (BHV1), Bovine respiratory syncytial virus (BRSV), and Bovine Parainfluenza type 3 virus (BPIV); cytokine expression in peripheral white blood cells, and BHV1-specific IgA titers in nasal secretions following the vaccination. A total of 60 calves (1 month old) were administered an IN MLV vaccine containing BHV1, BRSV, BPIV (Inforce 3) and randomly assigned to one of two experimental groups: ITM (n = 30; Multimin90, containing Se, Cu, Zn, and Mn) or SAL (n = 30; sterile saline). There was a consistent decay in virus-specific SNA titers in both groups.

Objectives: To study neutrophil gelatinase-associated lipocalin (NGAL) levels in peripheral blood in SLE, and to propose a mechanism by which neutrophils secrete NGAL on stimulation with immune complexes (IC).
Methods: NGAL was measured by ELISA in two independent Swedish SLE cohorts acting as exploratory and validation cohort (n=124 and n=308, respectively), disease controls (n=38) and healthy controls (n=77). NGAL levels were measured in supernatant from IC-stimulated neutrophils in the presence or absence of a toll-like receptor 8 inhibitor (TLR8i).
Objective: Our study aimed to investigate the therapeutic effects of the Kuntai capsule in improving ovarian function in rats with transplantation of cryopreserved ovary.
Methods: Two mice ovary cell lines were cultured with Kuntai capsule decoction, and cell apoptosis was detected by MTT assay. A total of 90 SPF Sprague Dawley rats were included in this study.

Selective breeding is a potent method for developing strains with enhanced traits. This study compared the growth performance and stress responses of the genetically improved Abbassa Nile tilapia strain (G9; GIANT-G9) with a local commercial strain over 12 weeks, followed by exposure to stressors including high ammonia (10 mg TAN/L), elevated temperature (37 °C), and both for three days. The GIANT-G9 showed superior growth, including greater weight gain, final weight, length gain, specific growth rate, and protein efficiency ratio, as well as a lower feed conversion ratio and condition factor compared to the commercial strain.
