In the small domesticated ruminants the nucleus of the hypoglossal nerve is situated in close relationship to the median line in the middle of the elongated medulla. The nucleus is divided by the obex into a rostral and a caudal portion. In the sheep, four distinct subnuclei can be recognized, whereas in the dwarf goat great variations in the arrangement of cell groups exists. In both animals large and medium sized neurons are observed. The large neurons are characterized by densely packed small organelles. The medium sized neurons vary between a pale and an electron dense type. According to the size and morphology of the vesicles three types of synaptic contacts can be differentiated. Synapses of type 2 contain clear round and flattened vesicles and are partially accompanied by subsurface cisterns.

Background: Hypertension is characterized by upregulation of the renin-angiotensin system, increased blood-brain barrier (BBB) permeability, microglia activation within autonomic nuclei, and an intense sympathoexcitation. There is no information on the interplay of these events during the development of neurogenic hypertension. We sought to identify the interaction and time-course changes of Ang II availability, barrier dysfunction, microglia activation, and autonomic imbalance within autonomic areas during the development of neurogenic hypertension.

Patients with amyotrophic lateral sclerosis (ALS) do not develop oculomotor disturbances and vesicorectal dysfunction until end-stage disease owing to the survival of certain motor neurons (MNs), including oculomotor neurons and MNs within Onuf's nucleus. In sporadic ALS, adenosine deaminase acting on RNA 2 (ADAR2)-mediated editing of GluA2 mRNA at the Q/R site is compromised in lower MNs. We previously developed genetically modified mice with a conditional knockout of ADAR2 in cholinergic neurons (ADAR2/VAChT-Cre, Fast; AR2).

Obstructive sleep apnea (OSA) incurs a huge individual, societal, and economic burden. Specific and selective targeting of hypoglossal motor neurons could be an effective means to treat OSA. Bioluminescent-optogenetics (BL-OG) is a novel genetic regulatory approach in which luminopsins, fusion proteins of light-generating luciferase and light-sensing ion channels, increase neuronal excitability when exposed to a suitable substrate.
