Background/objectives: DNA vaccines are rapidly produced and adaptable to different pathogens, but they face considerable challenges regarding stability and delivery to the cellular target. Thus, effective delivery methods are essential for the success of these vaccines. Advancements in Raman light sheet microscopy have provided a powerful, non-invasive, marker-free method for imaging complex 3D biological structures, such as cell cultures and spheroids. By combining 3D tomograms made by Rayleigh scattering, Raman scattering, and fluorescence detection, this modality captures complementary spatial and molecular data, critical for biomedical research, histology, and drug discovery. Despite its capabilities, Raman light sheet microscopy faces inherent limitations, including low signal intensity, high noise levels, and restricted spatial resolution, which impede the visualization of fine subcellular structures.

The combination of macroporous cryogels with synthetic peptide factors represents a promising but poorly explored strategy for the development of extracellular matrix (ECM)-mimicking scaffolds for peripheral nerve (PN) repair. In this study, IKVAV peptide was functionalized with terminal lysine residues to allow its in situ cross-linking with gelatin macromer, resulting in the formation of IKVAV-containing proteinaceous cryogels. The controllable inclusion and distribution of the peptide molecules within the scaffold was verified using a fluorescently labelled peptide counterpart.
