Article Abstract

Bioassay-guided fractionation of the crude extract of Mitrephora thorelii (Annonaceae) led to the isolation of two clerodane-type diterpenes. Their structures were characterised on the basis of spectroscopic methods as 6alpha,16,18-trihydroxycleroda-3(4),13(14)-dien-15,16-olide (1) and 16-hydroxycleroda-3(4),13(14)-dien-15,16-olide (2). Compound 1 is a new compound. Compounds 1 and 2 exhibited inhibitory activity against the proliferation of human hepatoma BEL-7402 cells in vitro. Compound 2 also showed an in vivo anti-tumour effect against the growth of hepatoma H22 in mice.

Three butanolides derivatives, grewiifolides A-C, and nine clerodane diterpenes, grewiifolins M-U, as well as a known sterol were isolated from the twigs of Casearia grewiifolia. The chemical structures and configurations of all isolates were established by various spectroscopic means and chemical derivatization. In a cell-based phenotypic assay using the adipogenesis model of human bone marrow mesenchymal stem cells (hBM-MSCs), grewiifolide B significantly promoted adiponectin-secretion with EC value of 24.

Phytochemical investigation of the MeOH extract of needles led to the isolation and identification of a new clerodane-type diterpene, pinuseldarone (), along with a known flavonoid, 5,4'-dihydroxy-3,7,8-trimethoxy-6-C-methylflavone (), through HPLC purification. The structure of the new compound was elucidated using spectroscopic methods, including 1D and 2D NMR, as well as HRESIMS. Its absolute configuration was established through NOESY analysis and computational methods, including electronic circular dichroism (ECD) calculations and gauge-including atomic orbital NMR chemical shift calculations, followed by DP4+ probability analysis.

More than 20 natural products have been reported to modulate PCSK9-mediated cholesterol regulation, and small-molecule-derived proprotein convertase subtilisin/kexin type 9 (PCSK9) inhibitors continue to be developed and identified. Here, twelve undescribed clerodane-type diterpenes (1-9 and 12-14) and two known compounds were isolated from the chloroform-soluble extract of the dried fruits of Casearia grewiifolia Vent. using a PCSK9 mRNA expression monitoring assay.

Article Synopsis
  • Nine new clerodane-type diterpenoids (caseabalanspenes A-I) were discovered from the twigs and leaves of Casearia velutina, alongside six known compounds.
  • The structures of these compounds were identified using spectroscopic techniques and their molecular formulas were confirmed through high-resolution mass spectrometry.
  • Among the new compounds, caseabalanspene 3 showed significant anti-inflammasome activity, reducing LDH levels in a dose-dependent manner with an IC value of 2.90 μM.
