Background: Calcineurin, a protein involved in functions such as synaptic plasticity and neuronal survival, plays an important role in the pathophysiology of Alzheimer's disease. This study, randomized, investigated the effects of FK506 (FK), a calcineurin inhibitor, on the behavioral, histological, and biochemical alterations observed in models of neurotoxicity induced by NMDA or Aβ and in a transgenic model for AD, in addition to the organotypic culture model stimulated with NMDA.
Methods: This study involved models for AD to experiment injecting NMDA or Aβ1-42 into the hippocampus of male C57Bl/6 mice aged 8-12 weeks to induce a neurotoxicity model, treating double-transgenic APP/PS1 mice, expressing both mouse/human APP and mutant human PS1, with chronic FK506 for AD, in which, to enable NMDA or Aβ1-42 microinjections, another experiment including a stereotaxic surgery was performed on the C57Bl/6 mice.

Purpose: Tacrolimus could induce hepatotoxicity during clinical use, and the mechanism was still unclear, which posed new challenge for the prevention and treatment of tacrolimus-induced hepatotoxicity. The aim of this study was to investigate the mechanism of tacrolimus-induced hepatotoxicity and provide reference for drug development target.
Methods: In this study, biochemical analysis, pathological staining, immunofluorescent staining, immunohistochemical staining, transcriptomic analysis, Western blotting was used to investigate the mechanism of tacrolimus-induced hepatotoxicity in gene knockout mice and Wistar rats.

There is insufficient evidence comparing the outcomes of tacrolimus-based remission induction therapy with infliximab in refractory ulcerative colitis (UC) and evidence regarding optimal strategies after tacrolimus-based remission induction therapy. We conducted a multi-institutional retrospective study of patients with UC treated with tacrolimus or infliximab between January 2010 and March 2019. The proportion of clinical remission at week 8 and cumulative colectomy-free rate were examined using propensity score matching analysis.

Background: This study aims to investigate adverse drug reaction signals associated with coagulopathies through data mining using the Adverse Event Reporting System (FAERS) of the US Food and Drug Administration. Prompt identification of high-risk drugs provides a valuable basis for enhancing clinical drug safety.
Methods: The adverse event reports related to coagulopathies from Q1 2004 to Q2 2024 were extracted from the ASCII data packages in FAERS.
