Polycystic ovary syndrome (PCOS) is one of the most common, yet heterogeneous and complex, endocrine disorders in women of reproductive age. Although the aetiology of PCOS remains uncertain, emerging evidence has indicated that exposure of the female fetus to the hyperandrogenism milieu in utero may result in PCOS phenotype after birth. Such a phenomenon has been formulated as the fetal origin of PCOS, which intends to give a possible explanation for PCOS aetiology. Given that the epigenetic modifications are usually involved in the development and inheritance of many adult diseases with fetal origin, we propose a hypothesis here referred to as "epigenetic abnormality underlying the fetal origin of PCOS". It states that in utero hyperandrogenism exposure may disturb the epigenetic reprogramming in fetal reproductive tissue, thereby resulting in postnatal POCS phenotype in women of reproductive age. Meanwhile, the incomplete erasure of such epigenetic abnormality in germ cells after fertilization may promote the transgenerational inherence of POCS. Thus, this epigenetic abnormality hypothesis has established a novel mechanism for PCOS development and inheritance. If verified, our hypothesis would open new avenues for the possible intervention at the critical period of prenatal life to prevent PCOS development and inheritance in adult women. Moreover, analysis of the epigenetic phenotypes and identification of specific epigenetic changes may help develop new tools for monitoring fetal development under an in utero hyperandrogenism environment.

Fibroepithelial polyps are rare benign tumors originating from the mesoderm and are more commonly found in the renal pelvis and distal ureter and less frequently in the proximal ureter or bladder. This case report presents a fibroepithelial polyp occurring in the bladder of the fetus, showcasing its two-dimensional ultrasound, three-dimensional ultrasound, color Doppler, and spectral Doppler ultrasound findings, providing a reference for the accurate diagnosis of this condition.

Neuroblastoma is a rare tumour originating from neural crest cells, primarily occurring in the adrenal glands and sympathetic ganglia, with prenatal diagnosis often complicated by the difficulty in distinguishing it from other foetal abdominal or paraspinal masses. We present a case of foetal neuroblastoma in a 26-year old woman who, at 36 weeks of gestation, experienced absent foetal movements, leading to ultrasound confirmation of foetal demise with associated effusions. An emergency caesarean section revealed a stillborn male foetus with a previously undetected encapsulated mass in the posterior mediastinum, which was confirmed as neuroblastoma through histopathological analysis.

The germ-cell lineage ensures the creation of new individuals, perpetuating/diversifying the genetic and epigenetic information across the generations. We have been investigating the mechanism for germ-cell development, and have shown that mouse embryonic stem cells (mESCs)/induced pluripotent stem cells (miPSCs) are induced into primordial germ cell-like cells (mPGCLCs) with a robust capacity both for spermatogenesis and oogenesis and for contributing to offspring. These works have served as a basis for elucidating key mechanisms during germ-cell development such as epigenetic reprogramming, sex determination, meiotic entry, and nucleome programming.

A pyruvate dehydrogenase complex deficiency causes a reduction in adenosine triphosphate production and energy insufficiency, leading to neurological disorders. An abnormal E1-alpha protein originating from the gene with pathogenic variants is unable to communicate with E1-beta for the formation of the E1 enzyme, decreasing pyruvate dehydrogenase complex activity. In this study, we report a Vietnamese boy with lethargy, severe metabolic acidosis, increased serum lactate, hyperalaninemia, lactic acidosis, and globus pallidus lesions.

Background And Objectives: Maternal western-style diets that are high in glucose and fat have well-known cardiovascular effects on offspring, yet the combined influence of such diets during pregnancy is relatively less comprehended. This study investigates the impact of maternal high glucose and fat diet (HGF) on vascular constriction in offspring and the underlying mechanisms.
Methods And Results: Pregnant Sprague-Dawley rats were provided with either HGF or control diets.
