Primary intraocular lymphoma (PIOL) is a type of diffuse large B-cell lymphoma (DLBCL) that is related to primary central nervous system lymphoma (PCNSL). Whether their pathogenesis is similar is presently unknown. Immunoglobulin heavy chain variable genes (IGHV@) somatic mutations were analysed in five patients with PIOL, one patient with concomitant PCNSL and one patient with systemic DLBCL involving the eye. Six in-frame mutated clonal IGHV@ rearrangements were cloned from PIOL specimens. The sequences showed no evidence of antigen selection and revealed absence of intraclonal heterogeneity in four of five cases, suggesting that PIOL and PCNSL may differ in their ontogeny.

The multipotent stem cells of our body have been largely harnessed in biotherapeutics. However, as they are derived from multiple anatomical sources, from different tissues, human mesenchymal stem cells (hMSCs) are a heterogeneous population showing ambiguity in their in vitro behavior. Intra-clonal population heterogeneity has also been identified and pre-clinical mechanistic studies suggest that these cumulatively depreciate the therapeutic effects of hMSC transplantation.

Follicular lymphoma (FL) course is highly variable, making its clinical management challenging. In this incurable and recurring pathology, the interval between relapses tends to decrease while aggressiveness increases, sometimes resulting in the transformation to higher-grade lymphoma. These evolutions are particularly difficult to anticipate, resulting from complex clonal evolutions where multiple subclones compete and thrive due to their capacity to proliferate and resist therapies.

Waldenström macroglobulinemia (WM) is characterized by the expansion of clonal lymphoplasmacytic cells; the MYD88L265P somatic mutation is found in >90% of patients, but malignant B cells may still display intra-clonal heterogeneity. To assess clonal heterogeneity in WM, we generated and performed single-cell RNA sequencing of CD19 sorted cells from five patients with and two patients with genotype as well as two healthy donors. We identified distinct transcriptional patterns in the clonal subpopulations not only between the two genetically distinct WM subgroups but also among patients, which affected the B cell composition in the different subgroups.
