Context: Active placental transport of maternal iodide by the thyroidal sodium iodide symporter (NIS) provides an essential substrate for fetal thyroid hormone synthesis. NIS is expressed in trophoblast and is regulated by human choriogonadotropin (hCG). In thyroid, iodide down-regulates expression of several genes including NIS. Placentas of iodine-deficient rats demonstrate up-regulation of NIS mRNA, suggesting a role for iodide in regulating placental NIS.

Objectives And Methods: The objectives were to examine effects of iodide on expression of NIS and hCG in BeWo choriocarcinoma cells. Gene expression was studied by quantitative real-time PCR. Effects on NIS protein expression were assessed by Western blotting. Functional activity of NIS was measured by (125)I uptake. Expression of hCG protein was assessed by immunoassay of secreted hormone.

Results: Iodide inhibited NIS mRNA and membrane protein expression as well as (125)I uptake, which were paralleled by decreased betahCG mRNA expression and protein secretion. Iodide had no effects on pendrin expression. Addition of hCG increased NIS mRNA expression. This effect was partially inhibited by addition of iodide. The inhibitory effects of iodide on NIS mRNA expression were abolished by propylthiouracil and dithiothreitol.

Conclusions: We conclude that expression of placental NIS is modulated by maternal iodide. This may occur through modulation of hCG effects on NIS and hCG gene expression.

Background: Maternal hypertensive disorders of pregnancy (HDP) was associated with increased risk of congenital hypothyroidism in preterm infants, but its underlying mechanisms remain unclear.

Objective: To investigate the possible mechanisms by which intrauterine exposure to HDP affects thyroid hormone synthesis in preterm infant rats.

Methods: preterm infant rats were obtained by Caesarean section delivery from the L-NAME group and Control groups which was induced by L-NAME and saline, respectively.

The toxicological role of Myricetin in the progression of human anaplastic thyroid cancer SW1736 cell line.

Cellular and Molecular Endocrine Research Center, Research Institute for Endocrine Sciences, Shahid Beheshti University of Medical Sciences, Tehran, Iran. Electronic address:

Aims And Background: Anaplastic thyroid cancer cells lack the capacity to effectively accumulate iodine and are therefore unresponsive to treatment with radioactive iodine. The main objective of this study was to examine the possible therapeutic effects of Myricetin on the SW1736 ATC cell line. In this study, we assessed the influence of Myricetin on iodide absorption, sodium iodide symporter gene expression, and apoptosis induction.

Background: Resveratrol, a herbal phytoalexin, is known to have anti-tumor effects in several tumors including thyroid cancer cells.

Aim: The aim of this study was to determine the effects of resveratrol on the expression of BRAF, ERK and NIS mRNA levels and protein expression in B-CPAP human thyroid papillary cancer cell line.

Methods: B-CPAP cells were treated with resveratrol at concentrations of 10-100 μM for 24-48-72 h.

Article Synopsis
  • This study examines the role of STAT6, a key transcription factor, in regulating the sodium/iodide symporter (NIS), which is crucial for thyroid hormone production.* -
  • Findings indicate that in Graves' disease, the thyroid-specific antibody (TSAb) increases NIS levels through STAT6 signaling, and blocking STAT6 can reduce NIS expression and improve thyroid function.* -
  • The research suggests that targeting STAT6 could provide new treatment options for Graves' disease by modulating NIS expression and thyroid activity.*

Article Synopsis
  • The study used the National Immunization Survey data and regression tree methodology to analyze how demographic and geographic factors affected COVID-19 vaccination rates over time, revealing previously unrecognized patterns.
  • Three models were created to track the likelihood of individuals receiving the first vaccine dose, completing the vaccination series, and getting a booster dose, with specific demographic groups showing different propensities based on age, recent vaccinations, and work or school requirements.
  • Key findings included that older adults were more likely to get vaccinated and boosted, individuals who had received other vaccines recently were more likely to start and complete their vaccinations, and those with vaccination requirements from work or school were also more inclined to complete the primary series.
