Atrial fibrillation (AF) is the most common arrhythmia in clinical practices. Mediated by muscarinic type 2 receptors (M(2)Rs), acetylcholine affects electrophysiological activities of atrial myocytes and may contribute to the onset of AF. In order to characterize the distribution of M(2)Rs in the atrial myocardium, different atrial regions in both the SD rat and human were dissected. Atrial myocytes were isolated with type II collagenase. The M(2)Rs expression in these atrial tissues and myocytes was detected by immunofluorescent staining and confocal laser scanning biological microscope. The results showed the highest density of M(2)Rs in atrial myocytes of the left atrial posterior wall. It is concluded that there is a marked spatial heterogeneity in the expression of the M(2)Rs in the atrium, which might create a substrate that would favor the initiation and maintenance of acetylcholine-induced AF.

Retinoic acid signaling pathway plays a role in regulating vertebrate development, cell differentiation, and homeostasis. As a key enzyme that catalyzes the oxidation of retinal to retinoic acid, aldehyde dehydrogenase 1 family member A2 (Aldh1a2) is involved in cardiac development, while whether it functions in heart diseases remains to be studied. The effect of a citrus-derived flavonoid, hesperetin, on the automaticity and contraction of isolated guinea pig myocardium was examined. Hesperetin inhibited the rate of ectopic action potential firing of the pulmonary vein myocardium; the slope of the diastolic depolarization was decreased with minimum change in the action potential waveform. The effect was dependent on the concentration; the EC value for firing rate was 56.

Nestin-expressing hair-follicle-associated pluripotent (HAP) stem cells from mouse and human have been shown to differentiate into neurons, glia, keratinocytes, smooth muscle cells, cardiac muscle cells, and melanocytes in vitro. HAP stem cells have promoted the recovery of peripheral nerve and spinal cord injuries in mouse models by differentiating into glial fibrillary acidic protein (GFAP)-positive Schwann cells. HAP stem cells enclosed on polyvinylidene fluoride membranes (PFM) were transplanted into the severed thoracic spinal cord of nude mice.
