The filamentous ascomycete Sordaria macrospora accumulates melanin during sexual development. The four melanin biosynthesis genes pks, teh, sdh and tih were isolated and their homology to genes involved in 1,8 dihydroxynaphthalene (DHN) melanin biosynthesis was shown. The presence of DHN melanin in S. macrospora was further confirmed by disrupting the pks gene encoding a putative polyketide synthase and by RNA interference-mediated silencing of the sdh gene encoding a putative scytalone dehydratase. Because melanin occurs in fruiting bodies that develop through several intermediate stages within 7 days of growth, a Northern analysis of a developmental time-course was conducted. These data revealed a time-dependent regulation of teh and sdh transcript levels. Comparing the transcriptional expression by real-time PCR of melanin biosynthesis genes in the wild type under conditions allowing or repressing sexual development, a significant downregulation during vegetative growth was detected. Quantitative real-time PCR and Northern blot analysis of melanin biosynthesis gene expression in different developmental mutants confirmed that melanin biosynthesis is linked to fruiting body development and is under the control of specific regulatory genes that participate in sexual differentiation.

This article presents new data on the integrated use of colloidal solutions of nanoparticles and low-intensity laser radiation on the biosynthetic activity of the medicinal mushroom . Traditional mycological methods, colloidal solutions of biogenic metals, and unique photobiological methods have also been used. It was found that colloidal solutions of nanoparticles of all metals used increased the growth characteristics of (55-60%), while irradiation of the fungal inoculum with laser light in a medium with nanoparticles reduced the growth activity of mycelia by 12.

Over the past ten years, there has been an increasing demand for reliable consumer wearables as users are inclined to monitor their health and fitness metrics in real-time, especially since the COVID-19 pandemic. Reflectance pulse oximeters in fitness trackers and smartwatches provide convenient, non-invasive SpO measurements but face challenges in achieving medical-grade accuracy, particularly due to difficulties in capturing physiological signals, which may be affected by skin pigmentation. Hence, this study sets out to investigate the influence of skin pigmentation, particularly in individuals with darker skin, on the accuracy and reliability of SpO measurement in consumer wearables that utilise reflectance pulse oximeters.

The effect of skin pigmentation on photoplethysmography and, specifically, pulse oximetry has recently received a significant amount of attention amongst researchers, especially since the COVID-19 pandemic. With most computational studies observing overestimation of arterial oxygen saturation (SpO) in individuals with darker skin, this study seeks to further investigate the root causes of these discrepancies.
