Mastoparans, a group of amphiphilic tetradecapeptides, are the major peptides in social wasp venoms and possess a variety of biological activities. Here we report the first crystal structure of mastoparan from Polistes jadwagae (MP-PJ) at 1.2 A resolution. The crystals belong to the space group P2(1) with eight molecules in an asymmetric unit. In contrast to the previous observations that the alpha-helical conformation only exists in the membrane-bound state of mastoparans, all of the MP-PJ molecules are in possession of the alpha-helical conformation even in the absence of trifluorethanol or detergents in the crystallization system. The high-resolution structure enables us to compare the conformation differences of MP-PJ with NMR results of other mastoparans. Together with biochemical results, we propose that the interactions between mastoparan molecules play an important role in forming the alpha-helical conformation, which is highly related to their biological activities.

Peptide stapling is an effective strategy to stabilise α-helical peptides, enhancing their bioactive conformation and improving physiochemical properties. In this study, we apply our novel diyne-girder stapling approach to the MDM2/MDMX α-helical binding region of the p53 transactivation domain. By incorporation of an unnatural amino acid to create an optimal , + 7 bridge length, we developed a highly α-helical stapled peptide, 4, confirmed circular dichroism.

A systematic structure-activity and computational modeling analysis of a series of glucagon-like peptide-1 receptor (GLP-1R) agonists based upon an ultra-short GLP-1 peptide, H-His-Aib-Glu-Gly-Thr-Phe-Thr-Ser-Asp-Bip-Bip-NH2, was conducted. This highly potent 11-mer peptide led to a deeper understanding of the α-helical bias of strategic α-methylation within the linear parent template as well as optimization of GLP-1R agonist potency by 1000-fold. These data were correlated with previously reported co-structures of both full-length GLP-1 analogs and progenitor N-terminal GLP-1 fragment analogs related to such ultra-short GLP-1R agonist peptides.
