The results of copolymer characterization by coupling of chromatography and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) techniques and subsequent calculation of copolymer composition using a novel software tool 'MassChrom2D' are presented. For high-resolution mass analysis copolymer samples were fractionated by means of liquid adsorption chromatography (LAC). These fractions were investigated off-line by MALDI-TOF MS. Various mono-n-butyl ethers of polyethylene oxide-polypropylene oxide copolymers (PEO-co-PPO) were investigated. As well as the copolymer composition presented in two-dimensional plots, the applied approach can give additional hints on specific structure-dependent separation conditions in chromatography.

Rationale: Fentanyl and fentanyl analogs continue to pose a serious threat to the public health. The vast number of fentanyl analogs emerging on the black-market call for optimized analytical methods for the detection, analysis, and characterization of these extremely dangerous drugs.

Methods: Atmospheric pressure solids analysis probe (ASAP) mass spectrometry was used for the rapid analysis of 250 synthetic opioid standards, including 211 fentanyl analogs, 32 non-fentanyl related opioids, and 8 fentanyl precursors.

GDFold2: A fast and parallelizable protein folding environment with freely defined objective functions.

Protein Sci

February 2025

MOE Key Laboratory of Bioinformatics, School of Life Sciences, Tsinghua University, Beijing, China.

An important step of mainstream protein structure prediction is to model the 3D protein structure based on the predicted 2D inter-residue geometric information. This folding step has been integrated into a unified neural network to allow end-to-end training in state-of-the-art methods like AlphaFold2, but is separately implemented using the Rosetta folding environment in some traditional methods like trRosetta. Despite the inferiority in prediction accuracy, the conventional approach allows for the sampling of various protein conformations compatible with the predicted geometric constraints, partially capturing the dynamic information.

scHiGex: predicting single-cell gene expression based on single-cell Hi-C data.

NAR Genom Bioinform

March 2025

Department of Computer Science, University of Miami, Coral Gables, FL 33146, United States.

A novel biochemistry experiment named HiRES has been developed to capture both the chromosomal conformations and gene expression levels of individual single cells simultaneously. Nevertheless, when compared to the extensive volume of single-cell Hi-C data generated from individual cells, the number of datasets produced from this experiment remains limited in the scientific community. Hence, there is a requirement for a computational tool that can forecast the levels of gene expression in individual cells using single-cell Hi-C data from the same cells.

Recently, lithium-sulfur batteries have captivated those in the energy storage industry due to the low cost and high theoretical capacity of the sulfur cathode (1675 mA h g). However, to enhance the practical usability of Li-S batteries, it is crucial to address issues such as the insulating nature of sulfur cathodes and the high solubility of lithium polysulfides (LiPS, LiS , 4 ≤ ≤ 8) that cause poor active sulfur utilization. Designing innovative sulfur hosts can effectively overcome sulfur bottlenecks and achieve stable Li-sulfur batteries.
