Several materials were evaluated for potential use in a bioreactor system for a tissue-engineered cornea. Two types of cytotoxicity tests were performed using human corneal stromal fibroblasts: a 24h cytotoxicity test based on the ASTM standard F813-01 and a 7 days growth inhibition test. It was determined that culture configuration, autoclaving and materials surface preparation were all important factors influencing cell viability. Poly(etheretherketone) and titanium-6Al-4V were found to be the most appropriate materials for use in a corneal bioreactor system. Furthermore, poly(oxymethylene) copolymer and poly(tetrafluoroethlylene) are not safe for use with human corneal fibroblasts after autoclaving.

Considering the unique composition and structure of the cell wall of , we investigated the effects of CmpLs on L-glutamate synthesis and transport in SCgGC7, a constitutive L-glutamate efflux strain. This study emphasizes the critical importance of closely monitoring and controlling the sialic acid content in therapeutic glycoproteins, including EPO, interferon-γ, Orencia, Enbrel, and others, as the level of sialylation directly impacts their pharmacokinetics (PK), immunogenicity, potency, and overall clinical performance due to its influence on protein clearance via hepatic asialoglycoprotein receptors (ASGPR). The ASGPR recognizes and binds to glycoproteins exposed to terminal galactose or N-acetylgalactosamine residues, leading to receptor-mediated endocytosis. The possibility to produce recombinant adeno-associated virus (rAAV) by adherent HEK293T cells was studied in a stirred tank bioreactor (STR) culture of cell aggregates. A proof-of-concept of rAAV production was successfully demonstrated in a process where single cells were first expanded, then cell aggregates were formed by dilution into a different medium 1 day before triple plasmid transfection was conducted. An alternative approach for the STR inoculation using a seed taken from a high cell density perfusion (HCDP) culture was also investigated.
