Objective: To separate and identify the exosomes derived from a mouse hepatoma carcinoma cell line (H22) and to detect their protein composition, and to investigate the possibility of using these exosomes as a kind of tumor vaccine.

Methods: Exosomes were purified by serial ultracentrifugation and sugar density ultracentrifugation, and then they were observed and identified by electron microscopy. Exosomes underwent peptide mass fingerprint and Western blot analyses.

Results: H22 cell-derived exosomes were 20-90 nm round or oval vesicles. The exosomes expressed HSP70, ICAM-1, EF-G2, DLC-A, C-myc protein and Vav-2 protein.

Conclusion: Serial ultracentrifugation and sugar density ultracentrifugation can be used to purify H22 cell-derived exosomes. H22 cell-derived exosomes express a distinct set of proteins involving in and/or relating to antigen presentation (HSP70, ICAM-1), migration (DLC-A), adhesion (ICAM-1), cytoskeleton (EF-G2) and tumour antigens (C-myc, Vav-2). The exosomes have immunogenicity.

The lipopeptides produced by Bacillus subtilis have anti-cancer potential. We had previously identified a secondary metabolite of B. subtilis strain Z15 (BS-Z15), which has an operon that regulates lipopeptide synthesis, and also demonstrated that the fermentation products of this strain exerted antioxidant and pro-immune effects.

Immune checkpoint blockade (ICB) therapy, particularly antibodies targeting the programmed death receptor 1 (PD-1) and its ligand (PD-L1), has revolutionized cancer treatment. However, its efficacy as a standalone therapy remains limited. Although ICB therapy in combination with chemotherapy shows promising therapeutic responses, the challenge lies in amplifying chemotherapy-induced antitumor immunity effectively.

Objective: Diabetic hepatocellular carcinoma (HCC) patients have high mortality and metastasis rates. Diabetic conditions promote neutrophil extracellular traps (NETs) generation, which mediates HCC metastasis and invasion. However, whether and how diabetes-induced NETs trigger HCC invasion is largely unknown.

Cell-derived microparticles, which are recognized as nanosized phospholipid bilayer membrane vesicles, have exhibited great potential to serve as drug delivery systems in cancer therapy. However, for the purpose of comprehensive therapy, microparticles decorated with multiple therapeutic components are needed, but effective engineering strategies are limited and still remain enormous challenges. Herein, BiSe nanodots and doxorubicin hydrochloride (DOX) co-embedded tumor cell-derived microparticles (BiSe/DOX@MPs) are successfully constructed through ultraviolet light irradiation-induced budding of parent cells which are preloaded with BiSe nanodots and DOX via electroporation.
