A samarium-promoted cyclopropanation of unmasked alpha,beta-unsaturated acids is described. This reaction can be carried out on (E)- or (Z)-alpha,beta-unsaturated carboxylic acids. In all cases the process is completely stereospecific and stereoselective. A mechanism has been proposed to explain the cyclopropanation reaction.

We herein disclose the synthesis of a bicyclo [5,1,0]-octane ring system consisting of a cycloheptane core fused with a cyclopropane unit of naturally occurring sesquiterpene capillosanane V. Initially, an unsuccessful RCM reaction of a properly functionalized bis-olefinic precursor was attempted. Finally, an intramolecular Reformatsky reaction was employed to construct the cycloheptane core present in the target structure.

Asymmetric gem-Hydroboration and gem-Hydrogenation of Ynamides: A New Gateway to Chiral Fischer Carbene Complexes and their Catalytic Transformations.

Angew Chem Int Ed Engl

January 2025

Ynamides, when reacted with H2 or HBpin in the presence of [Cp*RuCl]4, convert into chiral-at-metal Fischer carbenes by regioselective gem-hydrogenation or gem-hydroboration of the polarized triple bond, respectively. gem-Hydroboration concomitantly affords a carbogenic borylated stereocenter adjacent to the ruthenium carbene unit, the configuration of which can be controlled using an Evans auxiliary. These are the first examples of asymmetric gem-addition reactions to alkynes known in the literature; representative pianostool ruthenium carbene complexes formed by this unconventional route were characterized by crystallographic and spectroscopic means.

A modular approach was developed for the first catalytic asymmetric total syntheses of naturally occurring C30 terpene quinone methides and their non-natural stereoisomers, which feature the presence of an unprecedented spiro[4.4]nonane-containing 6-6-6-5-5-3 hexacyclic skeleton. Resting on a chiral phosphinamide-catalyzed enantioselective reduction of 2,2-disubstituted cyclohexane-1,3-dione, a concise route for the synthesis of enantioenriched 6-6 bicyclic fragment was developed.

Herein, we report a Lewis acid-mediated ring expansion of donor-acceptor cyclopropanes (DACs) to substituted azetidines via nucleophilic nitrogen group transfer from readily accessible iminoiodinane. This protocol operates under mild, transition-metal-free conditions, and showcases excellent chemoselectivity, along with broad functional group tolerance. We report for the first time that challenging alkyl donor-acceptor cyclopropanes can undergo ring expansion leading to aliphatic azetidines without relying on external oxidants or precious transition-metal catalysts.

A turn-on fluorescent probe containing a β-ketoester moiety for the selective detection of intracellular hydrazine.

Fluorogenic probes containing the β-ketoester structure were developed for selective hydrazine detection. The probe equipped with a cyclopropane moiety, having reduced steric hindrance, showed a higher reaction rate than its dimethyl counterpart. In live cell imaging, the probe detects intracellular hydrazine with minimal cytotoxicity.
