Optimized methods for scRNA-seq and snRNA-seq of skeletal muscle stored in nucleic acid stabilizing preservative.

Commun Biol

January 2025

Center for Research on Genomics and Global Health, National Human Genome Research Institute, National Institutes of Health, Bethesda, MD, 20892, USA.

Single cell studies have transformed our understanding of cellular heterogeneity in disease but the need for fresh starting material can be an obstacle, especially in the context of international multicenter studies and archived tissue. We developed a protocol to obtain high-quality cells and nuclei from dissected human skeletal muscle archived in the preservative Allprotect® Tissue Reagent. After fluorescent imaging microscopy confirmed intact nuclei, we performed four protocol variations that compared sequencing metrics between cells and nuclei enriched by either filtering or flow cytometry sorting.

Hierarchically aligned heterogeneous core-sheath hydrogels.

Nat Commun

January 2025

Institute of Innovative Materials, Department of Chemistry, College of Science, Southern University of Science and Technology, Shenzhen, China.

Natural materials with highly oriented heterogeneous structures are often lightweight but strong, stiff but tough and durable. Such an integration of diverse incompatible mechanical properties is highly desired for man-made materials, especially weak hydrogels which are lack of high-precision structural design. Herein, we demonstrate the fabrication of hierarchically aligned heterogeneous hydrogels consisting of a compactly crosslinked sheath and an aligned porous core with alignments of nanofibrils at multi-scales by a sequential self-assembly assisted salting out method.

Electronic address:

Anti-mullerian hormone (AMH) detection receives much attention since it is used as an ideal biomarker for quantitative assessment of ovarian reserve. The present study proposed a first report on the use of MOF-on-MOF as an electrochemical sensor for recognizing AMH in buffer and serum media. The MOF-on-MOF, MIL-88 B@UiO66NH was synthesized by the internal extended growth method (IEGM) involving MIL-88 B on UiO66NH by in situ method for the first time.

Emergence as a phenomenon is embedded and expressed in the natural world, and in social systems. Introduced nearly 150 years ago in a philosophical context, it has since been applied in nearly every natural and social science. However all of these uses are not congruent, as the range of emergences in this Special Issue reflect as well; this has limited the accumulation of knowledge about emergence, as well as its development as a discipline.
