A fluorescence method for determining transport of charged compounds across lipid bilayer.

There is a constant need for simple, economical and time-efficient methods which allow evaluating a compound's ability to penetrate the biological membrane, one of the key parameters needed to characterize biologically active compounds. In the paper we propose a new method of permeability determination. Instead of detecting the compound's concentration directly, we employ an approach in which the membrane interface is labeled with a fluorescein lipid probe; the probe is sensitive to the presence of charged compounds. The fluorescence intensity changes of the dye permanently attached to both sides of a model lipid bilayer are measured. Specifically, the time course of the fluorescence intensity changes following a rapid induction of a non-equilibrium state of the sample allows the evaluation of the membrane permeability for the compound. The method was validated by the determination of the phenyltin compound's transport through the model phosphatidylcholine unilamellar liposome bilayer.

A novel copper and iron doped containing chitosan and heparin sodium carbon dots (CS-Cu,Fe/HS) nanozyme was formulated through a single-step microwave digestion method. CS-Cu,Fe/HS exhibits excellent peroxidase (POD)-like activity and positive charge characteristics, and it can oxidize the negatively charged 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) in the presence of HO to produce a green compound (ox-ABTS). Furthermore, CS-Cu,Fe/HS enhances electron transfer and provides additional active sites through the valence state transformations of Fe/Fe and Cu/Cu.

This article investigates the influence of dopant molecules on the structural and dynamic properties of lipid bilayers in liposomes, with a focus on the effects of dopant concentration, size, and introduced electric charge. Experimental studies were performed using electron paramagnetic resonance (EPR) spectroscopy with spin probes, complemented by Monte Carlo simulations. Liposomes, formed via lecithin sonication, were doped with compounds of varying concentrations and analyzed using EPR spectroscopy to assess changes in membrane rigidity.

Cisplatin-based platinum compounds are important clinical chemotherapeutic agents that participate in most tumor chemotherapy regimens. Through density-functional theory calculations, the formation and stability of the inorganic oxide carrier, the mechanisms of the hydrolysis reaction of the activated platinum compound, and its binding mechanism with DNA bases can be studied. The higher the oxidation state of Pt (II to IV), the more electrons transfer from the magnesia-gold composite material to the platinum compound.
