Unexpected reactivity of the 9-aminoacridine chromophore in guanidylation reactions.

The 9-aminoacridine chromophore is an important building block of DNA-targeted chemotherapeutic agents. The success of 1-[2-(acridin-9-ylamino)ethyl]-1,3-dimethylthiourea as a carrier group in cytotoxic platinum-intercalator conjugates prompted us to explore the synthesis of an analogous guanidine-functionalized acridine. In a successful effort to generate such a derivative, various methods of guanidylation were employed, which demonstrate that the acridine C9-N9 linkage is highly susceptible to electrophilic and nucleophilic attack. The newly established reactivities provide efficient pathways to novel cyclic and spirocyclic acridine derivatives.

Rationale: Liquid atmospheric pressure matrix-assisted laser desorption/ionisation (AP-MALDI) has been shown to enable the production of electrospray ionisation (ESI)-like multiply charged analyte ions with little sample consumption and long-lasting, robust ion yield for sensitive analysis by mass spectrometry (MS). The molecular interaction between hemoglobin (HHb), the major human heme protein, and the acridine dyes acridine orange (AO) and 9-aminoacridine (9AA) was studied by various spectroscopic, calorimetric and molecular modeling techniques. The dyes formed stable ground state complex with HHb as revealed from spectroscopic data. Temperature dependent fluorescence data showed the strength of the dye-protein complexation to be inversely proportional to temperature and the fluorescence quenching was static in nature.

An acridinium-based sensor as a fluorescent photoinduced electron transfer probe for proton detection modulated by anionic micelles.

A water-soluble fluorescent pH sensor of 9-amino-10-methylacridinium chromophore with the 2-(diethylamine)ethyl chain as a receptor shows an "off-on" response going from basic to acidic solution. Photoinduced electron transfer has been directly demonstrated to be the quenching mechanism by the observation of the long-lived acridinyl radical. The interaction of the protonated sensor with anionic micelles causes a significant increase in the detection sensitivity of pH.

Thioflavin T (ThT) is amyloid specific fluorescence dye possessing the properties of molecular rotor. We have shown that Thioflavin T forms complexes with non-peptide polyanions heparin, polyadenylate and polystyrene sulphonate by means of absorption spectroscopy. In the presence of chiral polyanions - heparin and polyadenylate - induced optical activity of ThT occurs whereas interaction with achiral polystyrene sulphonate (PSS) does not lead to production of induced circular dichroism signal.
