Passive avoidance conditioning is analyzed in a three-compartment apparatus comprising the central light compartment, the dark dangerous compartment in which the electric foot shock was delivered, and the dark safe compartment where the rats were not punished. The passive avoidance performance was characterized, in addition to the latent period duration, by the number of visits into the safe compartment. The experimental data indicate that the latency of the passive avoidance response and the safe compartment preference obey different laws. The latency depends linearly on the number of shocks, while neither the safe compartment preference nor its relation to the number of shocks was observed. Piracetam had no effect on the latency, but enhanced the number of visits into the safe compartment. It is concluded that this modified model may be useful in an analysis of nootropic effects of neuropsychotropic substances.

Background: 7-Hydroxymethotrexate (7-OHMTX) is the main metabolite in plasma following high-dose MTX (HD-MTX), which may result in activity and toxicity of the MTX. Moreover, 7-OHMTX could produce crystalline-like deposits within the renal tubules under acidic conditions or induce renal inflammation, oxidative stress, and cell apoptosis through various signaling pathways, ultimately leading to kidney damage. The objectives of this study were thus to explore the exposure-safety relationship of two compounds and search the most reliable marker for predicting HDMTX nephrotoxicity.

The applicant subsequently proposed a new method of analyses, incorporating a tracer in the composition of a premix, which has been evaluated by EURL and recommended as official method for the quantification of the organic chromium content in premixtures and feedingstuffs. In view of the above, the Commission asked EFSA to issue a new opinion on the safety and efficacy of chromium proprionate (KemTRACE™ Chromium) as feed additive for all growing poultry species including the newly submitted information by the applicant that involves the use of the microtracer.
